Micromeria cilicica ve M. Juliana türlerinde antioksidan bileşiklerin HPLC ile analizi ve yapılarının aydınlatılması

Abstract
Bu doktora tez çalışmasında, Micromeria cilicica Hausskn. ex P.H. Davis (endemik) ve M. juliana (L.) Bentham ex Reichb. türlerinin topraküstü kısımlarından hazırlanan petrol eteri, aseton ve metanol ekstrelerinin toplam fenolik miktarları pirokatekole, toplam flavonoit miktarları kersetine eşdeğer olarak tayin edildikten sonra antioksidan aktiviteleri ? -karoten renk açılım, DPPH serbest radikali giderim ve süperoksit anyon radikali giderim yöntemleri kullanılarak belirlendi. Yüksek antioksidan aktivite gösteren ekstreler fraksiyonlandırıldı, elde edilen fraksiyonların da antioksidan aktiviteleri incelendi. Aktif çıkan fraksiyonlardan kromatografik yöntemler kullanılarak yedi madde saflaştırıldı. M. cilicica'nın aseton ekstresinden ikisi yeni (izotimonin 4'-metil eter, piperiton 7-O- ? -D-glikozit), dördü bilinen (ursolik asit, sudachitin, izomukronulatol, rutin) ve M. juliana'nın aseton ekstresinden bilinen bir madde (sakkaroz) elde edildi. Saf maddelerin yapılarının belirlenmesinde spektroskopik yöntemler ve antioksidan aktivitelerinin belirlenmesinde ? -karoten renk açılım, DPPH serbest radikali giderim ile süperoksit anyon radikali giderim yöntemleri kullanıldı. Ayrıca M. cilicica'nın aseton ve M. juliana'nın petrol eteri ekstrelerinden antioksidan aktivite gösteren fraksiyonların GC ve GC/MS analizleri yapıldı. GC ve GC/MS analizleri sonucunda yirmi beş maddenin yapıları belirlendi.Bu tez çalışması ile M. cilicica'dan hazırlanan tüm ekstrelerin ve M. juliana'nın petrol eteri ile aseton ekstrelerinin antioksidan aktiviteleri ilk kez incelendi. Ursolik asit hariç diğer saf maddelerin Micromeria türlerinden ilk kez elde edildiği tespit edildi. Ursolik asit ve rutin dışındaki diğer saf maddelerin antioksidan aktiviteleri de yine ilk kez bu tez çalışmasında belirlendi. Rutin'in, ? -karoten renk açılım, DPPH serbest radikali giderim ve süperoksit anyon radikali giderim yöntemlerinin hepsinde en yüksek aktiviteyi gösterdiği saptandı.Anahtar Kelimeler : Micromeria cilicica, Micromeria juliana, Lamiaceae, Antioksidan Aktivite In this thesis, the aerial parts of Micromeria cilicica Hausskn. ex P.H. Davis (endemic) and M. juliana (L.) Bentham ex Reichb. were extracted with petroleum ether, acetone and methanol, successively. Total phenolic and total flavonoid contents of these extracts were determined as pyrocatechol and quercetin equivalents, respectively, and their antioxidant activity, was carried out by using three different methods; namely, ß-carotene bleaching method, DPPH free radical scavenging activity and superoxide anion radical scavenging activity assays. The extracts that exhibited high antioxidant activity were fractionated, and then the antioxidant activity of all fractions were also determined. Seven compounds were isolated from active fractions by using chromatographic methods. Two new (isothymonin 4'-methyl ether, piperitone 7-O- ? -D-glucoside) and four known (ursolic acid, sudachitin, isomucronulatol, rutin) compounds were obtained from the acetone extract of M. cilicica, and a known compound (saccharose) from the acetone extract of M. juliana. For structural elucidation of the isolated compounds, spectroscopic methods were used while for their antioxidant activity, ß-carotene bleaching method, DPPH free radical scavenging activity and superoxide anion radical scavenging activity assays were used. Furthermore, GC and GC/MS analyses of active nonpolar fractions from the acetone extract of M. cilicica and the petroleum ether extract of M. juliana were performed and the structure of twenty five compounds were elucidated.In this thesis, the antioxidant activity of all extracts of M. cilicica, the petroleum ether and the acetone extracts of M. juliana was determined for the first time. All compounds except ursolic acid were isolated for the first time from Micromeria species. The antioxidant activity of these compounds, except for ursolic acid and rutin, was also determined for the first time in this study.
