Phillyrea latifolia L. bitkisinin etken maddeleri ve zayıflamada kullanılışı üzerine farmakognozik araştırmalar

Abstract
Bu çalışmada halk arasında zayıflamada kullanılan Phillyrea latifolia L. (pırnal), bitkisinin etken maddeleri ve yüksek enerjili yem ile kilo aldırılan hayvanlardaki etkisi araştırılmıştır. Tipik bir Akdeniz makisi olan Phillyrea latifolia örnekleri İstanbul Ayazağa'dan toplanmıştır.Kurutulup toz edilen yapraklar etanol ile tüketilmiş, sırasıyla sıvı-sıvı ekstraksiyonu ile kloroform ve etil asetat ekstreleri hazırlanmıştır. Madde içeriği daha zengin olan EtOAc ekstresi vakum likit kromatografisi yöntemi ile fraksiyonlandırılmıştır. Fraksiyonlardaki maddeler kromatotron ve kromatografik yöntemlerle (İTK, KK) ayrılmıştır. Sephadex LH?20 sütunda saflaştırılmıştır. Fenolik bileşiklerden Luteolin?7-O-glikozit ve klorojenik asit elde edilip yapıları spektral yöntemlerle belirlenmiştir. Ayrıca verbaskozit, ß-hidroksiakteozit olduğu düşünülen fenilpropanoit bileşikler elde edilmiş, analizleri devam etmektedir.Bitkinin zayıflamadaki etkisi yüksek enerjili yem ile şişmanlatılan Wistar-albino sıçanlarda araştırılmıştır. Kilolu sıçanlardan; bir gruba yüksek enerjili yem ile birlikte Phillyrea latifolia yaprak infüzyonu (PLE) gavaj yoluyla uygulanırken (YEY+PLE), ikinci gruba sadece yüksek enerjili yem verilmiştir (YEY). Kontrol grubu (K) ise sadece standart pellet yemle beslenirken son gruba standart pellet yem ve PLE uygulanmıştır (K+PLE). Beş haftalık uygulamanın ardından hayvanların kanlarında bazı biyokimyasal parametreler incelenmiş, karaciğer ve ince bağırsak dokuları histolojik olarak değerlendirilmiştir.YEY+PLE ve YEY gruplarının karaciğer ve ince bağırsak histolojileri karşılaştırıldığında, PLE uygulaması hücresel yapıları değiştirmezken, hücre sayısında artışa ve glikokaliks kalınlaşmasına neden olmuştur. YEY grubunda kan glukoz değerlerinin arttığı, YEY+PLE grubunda ise kontrol grubuna benzer olduğu görülmüştür. LDL (Low Density Lipoprotein), YEY grubunda anlamlı olarak artmış, K ve K+PLE grubunda bir değişiklik olmamıştır. YEY+PLE'lerde ise LDL'nin düştüğü görülmüştür. HDL (High Density Lipoprotein) değerlerinde hem kontrollerde hemde YEY grubunda istatistiksel anlamlılık yok iken YEY+PLE grubunda artmıştır Total kolesterol değeri yüksek enerjili beslenme ile artarken, YEY+PLE grubunda azalmıştır. Kontrol hayvanlarında PLE uygulaması, total kolesterolü değiştirmemiştir. Trigliserid düzeyi de YEY+PLE grubunda, kontrol grubundaki değerin altına inmiştir. Yağ dokuda sentezlenen bir hormon olan Leptin, YEY ile artarken YEY+PLE grubunda azalmıştır. In this study, the determination of active compounds in the leaves of Phillyrea latifolia L. which is being used for weight loss and slimming in folk medicine and effects of the leaves of this plant on overweight rats fed on high calorie diet was investigated. Samples of Phillyrea latifolia L. which is one of the most typical species of evergreen and maquis formation in Mediterranean were collected from Istanbul Ayazaga.The dried and powdered leaves of the plant were extracted with EtOH. Chloroform and ethyl acetate extracts were prepared by using liquid-liquid extraction method. Compund rich EtOAc extract was fractionated using vacuum liquid chromatography method. Fractions were seperated with chromatotron and chromatographic methods (TLC, PC) and purified on Sephadex LH?20 column.As phenolic compounds Luteolin?7-O-glucoside and chlorogenic acid were isolated and their structures were identified by spectral methods. Also, the analyses are still being made on verbascoside and ß-hydroxyacteoside which were identified as phenylpropanoid compounds.The effect of the plant in weight loss was investigated on the over-weight rats (wistar albino) fed with high-energy pellet. Over-weight rats were rendered (1) the leaves of Phillyrea latifolia infusion (PLE) administered by gavage, addition to high calorie diet (YEY+PLE group), (2) high calorie diet (YEY group). The control group (K) was fed with a standard pellet food. It was given a standard pellet food with the PLE as the fourth group (K + PLE). After five weeks, some biochemical parameters were examined in the blood and tissue samples (liver, intestine) were evaluated histologically. When compared to liver and intestine tissues histology in the groups of YEY and YEY+PLE; although PLE administration does not affect the sub cellular structure, it caused the increment in the number of cells and thickening of glycocalyx in comparison with YEY and YEY+PLE. Feeding with YEY caused high blood glucose but YEY+PLE group rats had similar blood glucose levels with control group. LDL (Low Density Lipoprotein) level was significantly higher in the YEY group, K and K+PLE groups had same one. LDL was decreased in YEY+PLE group and no significant changes were observed in the HDL (High Density Lipoprotein) levels of control and YEY groups. Moreover, total cholesterol levels were increased by high calorie diet and PLE administration decreased this increment. Total cholesterol level did not change in the control group. Triglyceride levels were additional decreased in YEY+PLE groups compared to control. Leptin was higher in YEY group but PLE administration normalized this increment.
