Abstract
ÖZET Schizosaccharomyces pombe' de Timidilat Sentaz Geninin İzolasyonuna İlişkin Çalışmalar Schizosaccharomyces pombe' de, gen izolasyonunda komplemantasyon tekniğinin uygulanmasında kullanılabilecek dTMP oksotrofları bulunmadığı için timidilat sentaz (TS) enzimini şifreleyen genin izolasyonunda değişik bir yaklaşım olarak, PCR ile oluşturulan prob aracılığıyla koloni melezlemesinin kullanılabilirliği araştırıldı. Farklı organizmalara ait TS enzimlerinde yüksek derecede homoloji gösteren iki bölgeye ait amino asit dizilerinden gidilerek tasarlanan timidilat sentaza özel primerlerin kullanımıyla S. pombe genomik DNA sında yapılan PCR ile oluşturulan ve DIG ile işaretlenen prob bu organizmanın pUR19 plazmidinde kurulu genom kitaplığında TS geninin taranmasında kullanıldı. Koloni melezlemesinde işaretlenen kolonilere ait plazmidlere PCR uygulaması sonucunda beklenen boyuttaki (300-400 Baz çifti) ürünün oluşması bu primerlerin ve tekniğin, bu organizmada da işlediği izlenimini verdi. Ancak örneklerden seçilen birkaç tanesinde yapılan enzimatik kesimlerin sonucunda bazılarının boyutlarının plazmid vektörün boyutundan küçük olduğu gözlendi. Bu bulgular, TS a özel primerlerin vektör içinde de tanıma bölgesi olabileceğini düşündürdüğünden, kontrol amacıyla farklı kökenli genleri içeren pUR19 ve yabancı gen içermeyen değişik özelliklerdeki plazmid vektörlere PCR uygulandı. Sonuçta tüm örneklerde benzer PCR ürününün elde edilmesi S. pombe' nin TS geninin izolasyonu için böyle bir yaklaşımın, en azından çalışmada kullanılan plazmid vektörde kurulu bir genom kitaplığı ile başarıya ulaşamayacağı sonucunu ortaya koydu. VI SUMMARY Studies on the Isolation of Thymidylate Synthase Gene from Schizosaccharomyces pombe. Since the dTMP auxotrophs, used in complementation technic applications, for gene isolation, does not exist in Schizosaccharomyces pombe, as a different approach to isolate the gene encoding thymidylate synthase(TS) enzyme, the relevance of colony hybridization via probes formed by PCR, was investigated. By tracing the amino acid sequences of two highly homologous regions in TS genes belonging to different organisms, primers specific for TS are designed and these primers are used in the formation of probe by PCR in S.pombe genomic DNA. Then, DIG-labelled probe is used in the screening of TS gene in the genomic library constructed in the pUR19 plasmid of this organism. The formation of the product of the expected size (300-400base pairs) as a result of PCR application to labeled colonies in colony hybridization, leads to the idea that these primers and this technic is operational in this organism. However, it is observed that the digestion by restriction enzymes in some selected samples results in smaller sizes than that of the plasmid vector. Because these findings bring about the idea of the existance of a recognition site in the vector, PCR was applied for control purposes to p(JR19 containing genes from different origins and to different plasmid vectors without insert. As a conclusion, obtaining similar PCR product in all samples yields the impression that such an approach to the isolation of TS gene proves to be unsuccessful, at least by using a genomic library constructed in the plasmid vector used in this study.
