Abstract
ÖZET Doku Kültüründe Paklitaksel Uygulanmış Normal ve Tumoral Kökenli Hücrelerde Amifostin ve Sisteamin'in Sitoprotektif Etkileri Bu çalışmada, normal ve tumoral kökenli hücrelerde paklitaksel'in oluşturacağı sitotoksik etki saptanarak, amifostin ve sisteamin'in bu sitotoksisite üzerine sitoprotektif etkileri araştırılmıştır. Bu amaçla, tumoral hücre soyu olarak fare derialtı kökenü L hücreleri ile insan serviks karsinomu kökenli HeLa hücreleri, normal hücre soyu olarak da Embriyonik Fare Fibroblast hücreleri kullanılmıştır. Deneylerimizin sonuçları, çoğalma hızı, mitoz indeksi ve işaretlenme indeksi gibi hücre kinetiği parametreleri ile değerlendirilmiştir. Bu amaçla, paklitaksel'in 6 (ig/ml ve 12 ug/ml konsantrasyonları 1-10 gün süresince tüm hücre soylarına tek başlarına ve amifostin veya sisteamin ile kombine olarak uygulanmıştır. Amifostin ve sisteamin'in 1 ug/ml konsantrasyonlarda tek başlarına uygulandığı deney gruplarındaki sonuçlara göre, yukarıdaki parametreler açısından tüm hücre soylarında istatistiksel açıdan anlamlı bir etki saptanmamıştır. Buna karşılık, paklitaksel'in uygulanan her iki konsantrasyonu da tüm hücre soylarında zamana bağlı olarak artan bir sitotoksik etkiye sebep olmuştur. Paklitaksel'in sitotoksik etkisi, hücre çoğalmasındaki baskılanma, mitoz fazı birikimi (G2/M blokajı) ve buna bağlı olarak sentez fazmda düşme olarak ortaya çıkmıştır. Deneylerimizde, HeLa hücrelerinin paklitaksel'in bu etkisine karşı çok duyarlı olduğu, Embriyonik Fare Fibroblast hücrelerinin ise tumoral hücrelere oranla paklitaksel' e karşı daha dirençli oldukları saptanmıştır. Amifostin ve sisteamin'in tumoral ve normal hücrelerdeki sitoprotektif etkilerinin araştırıldığı ve bu amaçla paklitaksel ile kombine uygulandıkları deney gruplarında ise, paklitaksel'in sitotoksik etkisi, amifostin veya sisteamin ile kombinasyonlarının uygulandığı tumoral hücre soylarında aynen devam ederken, normal hücre soyunda ortadan kalkmıştır. Paklitaksel'in amifostin ve sisteamin ile kombinasyon sonuçları birbirleriyle karşılaştırıldığında, paklitaksel'in amifostin ile kombinasyonunda ortaya çıkan sitoprotektif etkinin, sisteamin ile kombinasyondan daha güçlü olduğu saptanmıştır. Sonuç olarak, değerlendirilen parametrelerin ışığında, amifostin'in paklitaksel'in sitotoksik etkisine karşı, tumoral hücrelerde bir koruma sağlamadığı, buna karşılık normal hücrelerde ise, seçici bir koruma oluşturduğu ortaya konmuştur. xnı SUMMARY Cytoprotective Effects of Amifostine and Cysteamine on Cultured Normal and Tumour Cells Treated with Paclitaxel In this study, the cytotoxic effect occured by paclitaxel in normal and tumour cells was established, and cytoprotective effects of amifostine and cysteamine against this cytotoxicity were examined. L cells of mouse subcutaneous origin and human cervix carcinoma-derived HeLa cells were employed as tumour cell lines in the present study, while Mouse Embryonic Fibroblast as normal cell line. Results of the experiments were evaluated with cell kinetic parameters such as growth rate, mitotic index and labelling index. In the experiments, paclitaxel concentrations of 6 and 12 ug/ml were applied to the whole cell lines for 1-10 days, both solely and in combination with amifostine and cysteamine. In view of above mentioned parameters, a statistically significant effect was not determined in the whole cell lines, as seen from the results of the test groups in which 1 ug/ml of amifostine and cysteamine concentrations were applied solely. In contrast, both of applied concentrations of paclitaxel caused a cytotoxic effect increasing time dependently. The cytotoxic effect of paclitaxel appeared as repression of cell growth, mitotic phase accumulation (G2/M blockage) and a subsequent decline of synthesis phase. It was observed from the experiments that HeLa cells are very sensitive to paclitaxel, whereas Mouse Embryonic Fibroblast cells are quite resistant to it relative to tumour cells. In the test groups treated with combined drugs to investigate cytoprotective effects of amifostine and cysteamine on normal and tumour cell lines paclitaxel continued its cytotoxic effect in tumour cells exposed to combined treatment, but this effect disappeared in normal cell üne. When the results of combined paclitaxel and amifostine are compared to those of combined paclitaxel and cysteamine it was established that cytoprotective effect of combined paclitaxel and amifostine is more powerful than that of combined paclitaxel and cysteamin. In conclusion, this study clarified in the light of evaluated parameters that amifostine does not give tumour cells protection against cytotoxic effect of paclitaxel, but it gives protection to normal cells selectively.
