Patates (Solanum tuberosum L.) çeşitlerinde polifenol oksidaz genleri üzerine çalışmalar

Abstract
ÖZET Bu çalışmada; patates (Soîanum tuberosum L. ev. Yayla Kızı) tuberlerinde kahverengileşmeden sorumlu olan polifenol oksidaz (PPO) enzimi sentezinin `antisens` RNA teknolojisi kullanılarak engellenmesi amaçlandı. Bu amaçla patates tuber ve sürgünlerinde total RNA izolasyonu gerçekleştirildi. Daha sonra PPO transkriptleri total RNA' dan PPO genine özgü primerin kullanıldığı RT-PCR tekniği ile çoğaltıldı. Çoğaltılan PPO gen ailesine ait 1.5 kb'hk parça pUC19 plazmiti içine sokuldu. Rekombinant plazmit E. coli DH5a hücrelerine aktarıldı. Daha sonra PPO geni pBI-121 plazmiti içine ters konumlu olarak sokuldu. E. coli DH5a hücreleri içinde bulunan rekombinant pBI-121 plazmiti `Triparental Mating` sistemi ile Agrobacterium tumefaciens LBA 4404 hücreleri içine aktarıldı. Ters konumlu PPO geni A. tumefaciens LBA 4404 hücreleri aracılığıyla yüzey sterilizasyonlan yapılmış olan patates tuber disklerine aktarıldı. Tuber diskleri Murashige ve Skoog seçici besiyerlerinde kontrollü bitki büyüme kabininde tutularak dört hafta aralıklarla alt kültürlemeleri yapıldı. Yaklaşık 6-8 hafta sonra bitkiciklerde mikrotuber oluşumları gözlendi. NPTII (Neomisinfosfotransferaz) geninin genoma girdiği PCR ve `Dot` melezleme teknikleri kullanılarak kanıtlandı. Bulgulara ek olarak, gen aktarılan mikrotuberlerde PPO enzimi aktivitesinin ve kahverengileşmenin de fenotipik olarak azaldığı belirlendi. IX SUMMARY In this study, it was aimed to prevent a synthesis of polyphenol oxidase (PPO) enzyme which is responsible from enzymatic browning in potato (Solarium tuberosum L. ev. Yayla Kıza) tubers by using antisense RNA technology. For this purpose, total RNA was isolated from potato tubers and sprouts. After that, PPO transcripts were amplified using specific primers for PPO genes by RT-PCR technique from total RNA. Amplified 1.5 kb DNA fragment belonging to the PPO gene family was ligated into the pUC19 plasmid. The recombinant plasmid was transferred into the competant E. coli DH5ot cells. The PPO gene was inserted in antisense orientation into the pBI-121 plasmid which then transferred to E. coli DH5a cells. Recombinant pBI-121 plasmid was transferred to the Agrobacterium tumefaciens LBA 4404 strains from E. coli DH5a cells by using `Triparental Mating` system. Antisense PPO gene was transformed to the surface-sterilized potato tuber discs via A. tumefaciens LBA 4404. Tuber discs were cultured on selective Murashige and Skoog medium in controlled plant growth cabinet and subcultured for intervals of 4 weeks. Integration of NPTII (Neomycinphosphotranferase) gene in plant genome was confirmed by using PCR and `Dot` hybridization techniques. In addition, it was determined that PPO enzyme activity and also phenotipically browning decreased in transgenic microtubers.
