Abstract
ÖZET LİPAZ ENZİMİ İLE ENANTİOMER HİDROKSİ ASİDLERİN SENTEZİ Bu çalışmada yüksek enantiomerik zenginlikte 2-,3-,13- mono hidroksi tetradekanoik asid ve metil ester izomerlerinin enantioseçimli porcine pankreas lipaz hidrolizi ile sentezi amaçlanmıştır. Başlangıç maddesi olarak rasem 2-hidroksi tetradekanoik asit metil esteri, Hell- Volhard-Zelinsky yöntemine göre, 3- ve 13- mono hidroksi tetradekanoik asid metil esterleri ise asetoasetester yöntemine göre elde edilen eşdeğerleri 3-keto ve 13-keto tetradekanoik asit metil esterlerinin NaBüt ile indirgenmeleri sonucu sentezlenmiştir. Rasemik hidroksi esterler söz konusu olan rasemik hidroksi asitlere dönüştürülmüşlerdir. Gerek rasem ester gerekse rasem asid izomerlerinden izomerik R ve S enantiomerlerin yapı tayininde yararlanılmıştır. Rasemik 2-,3-,13- mono hidroksi tetradekanoik asid metil esterler, porcine pankreas lipaz enzimi ile değişik tür karıştırma, ortam, mol oranlan, pH, sıcaklık ve süre şartlarında enantioseçimli olarak hidroliz edilmişlerdir. 15 ayrı deneme ile %100'e yakın enantiomerik saflık veren optimum şartlar saptanmıştır. Sentezlenen enantiomerlerin yapılan C, H, İR, 'H-NMR ve 'H-NMR-shift, kütle ve kiral HPLC ile aydınlatılmıştır. 3-Hidroksi izomerine ait saf R ve saf S enantiomer esterlerin elde edilmesi ilk kez gerçekleştirilmiştir ve çalışmada önerilen bu yöntem orjinaldir. Sözü geçen enantiomerlerin enantiomerik zenginlik dereceleri % 100'e yalandır. Aynı şartlarda 2-hidroksi izomerine ait sentezlenen asid ve ester enantiomerleri arasında ancak R-konfigürasyonundaki asid yüksek enantiomerik zenginliktedir. 13-Hidroksi izomeri için porcine pankreas lipazı aynı şartlarda enantioseçimlilik göstermemiştir. X SUMMARY SYNTHESIS OF ENANTIOMERIC HYDROXY ACIDS WITH LD7ASE In this study, it is aimed to synthesize 2-, 3-, and 13-monohydroxy tetradecanoic acids and ite methyl ester isomers in high enantiomeric excess via enantioselective porcine pancreas (PP) lipase. The reactant, racemic 2-hydroxy tetradecanoic acid methyl ester, was synthesized according to the Hell-Volhard-Zelinsky method. 3-Keto and 13-keto tetradecanoic acid methyl esters were obtained by acetoacetic ester method and were reduced with NaBIL» to corresponding racemic 3- and 13-hydroxy tetradecanoic acid methyl esters. Racemic hydroxy esters were converted into relevant hydroxy acids. Racemic acid isomers as well as racemic esters were used in structural determination of isomeric R and S enantiomers. Racemic 2-, 3-, and 13-monohydroxy tetradecanoic acid methyl esters were enantioselectively hydrolyzed by using porcine pancreas lipase, with varying values of stirring, medium, molar ratio, pH, temperature, and duration. The optimum conditions were determined yielding almost 100% enantiomeric excess with 15 separate experiments. Structures of the enantiomers synthesized were determined by C, H, IR, ^-NMR and ^-NMR shift, mass spectrometry, and chiral HPLC. The method exhibited in this study as well as pure R and pure S enantiomer esters belonging to 3-hydroxy isomer were synthesized for the first time. The enantiomeric excess of the mentioned enantiomers approximates to 100%. It was found that between the synthesized acid and ester enantiomers of the 2- hydroxy isomer in the same conditions, the acid of R-configuration only is of high enantiomeric excess. Porcine pancreas lipase was not enantioselective in the same conditions for 13- hydroxy isomer. XI
