Abstract
Pek çok kanser türünde olduğu gibi, beyin tümörlerinde de bazı stres proteinlerinin anlatımında artış olduğu bilinmektedir. Özellikle Hsp27 ve Hsp70'deki bu artış, kanser hücrelerini tedaviye daha dirençli hale getirmektedir. Son yıllarda 14-3-3 proteininin bazı formlarının da çeşitli kanser türlerinde benzer bir etki gösterdiği bulunmuş ve bu nedenle 14-3-3 proteinleri de stres proteini olarak kabul edilmişlerdir. Stres proteinlerinin anlatımının baskılanması tedavi sürecine olumlu katkı sağladığından, bu tip proteinler yeni ilaç geliştirme çalışmalarında hedef haline gelmiştir.Bazı antioksidanların Hsp anlatımını baskıladığı bilinmektedir. Bu bağlamda, kanser tedavisinde destekleyici olarak kullanılan Viscum album bitkisinin de benzer bir etki gösterme potansiyeli bulunmaktadır. Bu yarı parazit bitkinin, ıhlamur ağacı üzerinden toplanmış alttürü (V. album ssp. album) metanolde çözünen antioksidanlar açısından oldukça zengindir.Bu çalışmanın ana hedefi, V. album ssp. album metanol ekstresinin, beyin tümörleri modeli olarak kullanılan C6 sıçan glioma hücrelerindeki stres proteinlerinin anlatımı üzerine etkisini belirlemek ve bu bitkinin kanser tedavisinde kullanımına ilişkin moleküler deliller elde etmektir. Çalışmada öncelikle, beyin tümörlerinde anlatımı artan Hsp27, Hsp70 ve 14-3-3 stres proteinleri araştırılmıştır.Çalışmanın ilk aşamasında, metanol ekstresinin hücre canlılığı üzerindeki etkisi incelenmiştir. Sitotoksisite analizlerinin sonuçlarına göre, hücreler üzerinde herhangi bir toksik ya da proliferatif etki göstermeyen ekstre konsantrasyonunun 100 ?g/ml olduğu bulunmuş ve çalışmanın her aşamasında hücrelere, bu dozda ekstre uygulanmıştır. Ekstre uygulamasının çözünebilir protein profilini nasıl etkilediği 1- ve 2-boyutlu elektroforez (1- ve 2-DE) teknikleriyle ortaya konulmuştur. Tek boyutlu jellerde ayrıştırılmış proteinler, Western blot tekniği ile membrana aktarılarak, Hsp27, Hsp70 ve 14-3-3 proteinlerine özgü antikorlarla işleme sokulmuştur. Bu analiz sonuçlarına göre, Viscum album ekstresi yüksek sıcaklıkla indüklenen 14-3-3 ?, ? ve ? formlarının anlatımını etkilememekte, Hsp27 ile 14-3-3 ß ve ? formlarının anlatımlarını indirgemekte ve Hsp70 anlatımını indüklemektedir.2-DE ile ayrıştırılan proteinler, gümüş nitratla boyanarak jel görüntüleme sistemi aracılığıyla karşılaştırılmıştır. Değişik örneklerde bazı farklılıklar gösteren yaklaşık 700 spot belirlenmiştir. Çalışmalar hedef stres proteinlerine odaklanılarak sürdürülmüştür. MALDI-TOF MS analizi ile uyumlu boya ile boyanmış 2-D jellerden kesilen toplam 20 spottan 13 tanesi MASCOT veri tabanında eşleştirilmiştir. Bu 13 proteine ait peptid kütle parmakizi spektrumları, Mowse skorları ve olası dizi eşleşme sonuçları birlikte değerlendirildiğinde, ısı şoku ile indüklenen 5 proteinin sıçanlardaki Hsp70'lerle eşleştiği gözlenmiştir. Bu proteinler Grp75 prekürsörü, Hsp72-ps1 (Hsp70), Hsc70, Hsc71 ve mitokondriyel Grp75'tir. Kalitatif veriler sHSP ailesinin bir üyesi olan beta-kristalin A1'in de indüklendiğine işaret etse de, bu protein için elde edilen Mowse skoru oldukça düşüktür.Sonuç olarak, Viscum album metanol ekstresinin, beyin tümörlerinde anlatımı belirgin şekilde artan stres proteinlerinden özellikle Hsp27, 14-3-3 ? ß ve 14-3-3 ?'yı baskılamak amacıyla kullanılabilme potansiyeli olduğu anlaşılmıştır. Bununla birlikte Hsp70 indüksiyonu dikkate alınmalıdır.Bu çalışmada elde edilen veriler, Viscum album metanol ekstresinin kimyasal içeriği ve etkime mekanizmasına ilişkin yeni araştırmaların önünü açmıştır. It is known that expression of some stress proteins are elevated in brain tumors, as in many cancer types. Especially, the increase in the expression of Hsp27 and Hsp70 make the cancer cells more resistant to the treatment. In recent years, it has been found that some forms of 14-3-3 protein have also similar effect on various cancer types and therefore, 14-3-3 proteins are approved as stress proteins. This type of proteins become targets in new drug development studies since suppressing the expression of stress proteins supports the treatment process.It is known that some antioxidants suppress Hsp expression. In this sense, Viscum album that is used as a supplement in cancer treatment, might have a potential of showing a similar effect. The subspecies of this semiparasitic plant, which was collected from lime trees (V. album ssp. album) is very rich regard to methanol soluble antioxidants.The main purpose of this study is to determine the effect of the methanolic extract of V. album ssp. album on the C6 rat glioma cells, which are used as a model of brain tumors and to achieve some molecular evidences related to the usage of this plant in cancer treatment. Primarily, Hsp27, Hsp70 and 14-3-3 proteins, those expression levels are elevated in brain tumors were investigated in this study.In the first stage of the study, the effect of methanolic extract on the cell survival was examined. According to the results of cytotoxicity analysis, the concentration that show neither a cytotoxic nor a proliferative effect on the cells was found as 100 ?g/ml and this dose of extract was applied to the cells on every stage of the study. The effect of the extract application on the soluble protein profile was exhibited by using 1- and 2- dimensional electrophoresis (1- and 2-DE) techniques. The proteins separated on 1-D gels were transferred to membrane using Western blotting technique and were reacted with the antibodies that are specific to the Hsp27, Hsp70 and 14-3-3 proteins. According to the results of this analyses, Viscum album extract does not effect the expression levels of 14-3-3 ?, ? and ? forms that are induced by high temperature; reduces the expression levels of Hsp27 and 14-3-3 ß and ? forms, and induces the expression level of Hsp 70.The proteins separated with 2-DE, were stained with silver nitrate and compared by using gel documentation system. Nearly 700 spots were detected in different samples, with slight dissimilarities. Studies were carried on by focusing target stress proteins. Only the thirteen spots of total 20 spots cut from 2-D gels, stained by dyes compatible with MALDI-TOF MS analysis, were matched in MASCOT database. By the overall assessment of the results of the peptide mass fingerprint spectra, Mowse scores and probable sequence matches belonging to these 13 proteins, 5 proteins induced by heat shock corresponded to Hsp70s in rats. These proteins are, precursor of Grp75, Hsp72-ps1 (Hsp70), Hsc70, Hsc71 and mitochondrial Grp75. Qualitative data showed that beta-kristallin A1, a member of sHSP family, was also induced, but the Mowse score of this protein was quite low.In conclusion, it seems that methanolic extract of Viscum album might have a potential in using to suppress Hsp27, 14-3-3 ? ß and 14-3-3 ? stress proteins, those expression levels are highly elevated in brain tumors. However, Hsp70 induction should be considered.Data obtained in this study, will lead to new studies on the chemical composition and action mechanism of Viscum album methanol extract.
