Hücre kültürlerinde oksidatif stres sonucu oluşan DNA hasarları ve zeytinyağının koruyucu etkisi

Abstract
Reaktif oksijen türleri (ROT) proteinler, membran lipitleri ve DNA gibi birçok hücresel molekülle reaksiyona girip bu moleküllerin yapısını değiştirerek hasar oluşturabilir. DNA'da meydana gelen oksidatif hasarın mutasyon, kanser oluşumu ve yaşlanma ile doğrudan ilişkili olduğu bildirilmektedir.Bazı doğal ürünler (zeytinyağı, yeşil çay, ayçiçeği yağı vb.) antioksidan aktiviteye sahiptir. Özellikle zeytinyağı serbest radikal süpürücü aktiviteye sahip birçok fenolik bileşik içerir. Bununla beraber, zeytinyağının oksidatif DNA hasarları üzerine etkisi, gene özgü olarak bugüne kadar hiç araştırılmamıştır.Bu çalışmada, oksidatif stres altındaki HeLa hücrelerinde belirli DNA bölgelerinde [transkripsiyonel olarak aktif olan APEX (Apürinik/apirimidinik endonükleaz) ve inaktif olan ß-globin gen kümesi ve mtDNA'nın belli bir bölgesinde] meydana gelen DNA hasarları ve zeytinyağı fenolik fraksiyonunun bu hasarları önleyici etkisi gene-özgü ?Kantitatif Polimeraz Zincir Reaksiyonu? (?Quantitative Polymerase Chain Reaction?, QPCR) analizi ile belirlendi. Elde edilen veriler, mtDNA'nın nükleer DNA'ya göre oksidatif strese karşı daha hassas olduğunu ve zeytinyağı fenolik fraksiyonu (ZFF)'nun nDNA'daki oksidatif hasarı tamamen önlediğini gösterdi. ZFF'nun hem normal hem de oksidatif stres koşullarında hücre içi ROS miktarını belirgin düzeyde azalttığı saptandı. Apoptoz düzeyi kaspaz 9 enzim aktivitesinin ölçümüyle belirlendi ve ZFF'nun oksidatif stres tarafından indüklenen apoptotik hücre ölümünü engelleyici etkiye sahip olduğu saptandı. Ayrıca mtDNA'da hasara neden olan oksidatif stresin sitokrom c oksidaz enzim aktivitesini yaklaşık % 44 oranında azalttığı ve ZFF'nun bu enzim aktivitesi üzerinde bir etkisinin olmadığı belirlendi.Sonuç olarak, zeytinyağının HeLa hücrelerini oksidatif strese karşı koruduğu ve bu etkisini, hücre içi ROT miktarını düşürerek, nDNA'yı koruyarak ve oksidatif stresin apoptotik etkisini baskılayarak gerçekleştirdiği ortaya konuldu. Reactive oxygen species (ROS) can cause damages on many cellular molecules such as proteins, membrane lipids and DNA by reacting with them and changing the structure. The resulting oxidative damage to DNA, is proposed to be directly related with mutagenesis, carcinogenesis and aging.Some natural products (olive oil, green tea, sunflower oil etc.) have antioxidant activity. Especially, olive oil contains many phenolic compounds which potentially possess free radical scavenging activity. However, the effect of olive oil on gene specific oxidative DNA damage has not been investigated up to date.In this study, DNA damages in specific sequences under oxidative stress [transcriptionally active APEX (Apurinic/apyrimidinic endonuclease) gene, transcriptionally inactive ß-globin gen cluster and a mtDNA fragment] and protective effect of olive oil phenolic fraction on these damages were determined on HeLa cells with gene-specific ?Quantitative Polymerase Chain Reaction? (QPCR) analysis. The obtained data showed that mtDNA is more sensitive to oxidative stress than nuclear DNA and phenolic fraction of olive oil completely prevents the oxidative damage on nuclear DNA. Phenolic fraction of olive oil significantly decreased the intracellular ROT level both in normal and oxidative stress conditions. Apoptosis was determined by the measurement of caspase 9 enzyme activity and it was observed that the phenolic fraction of olive oil has prohibitive effect on apoptotic cell death induced by oxidative stress. Furthermore, it was determined that the oxidative stress which causes damages on mtDNA, decreased the cytochrome c oxidase enzyme activity by approximately 44 % and observed that phenolic fraction of olive oil has not effect on this enzyme activity.As a result, it is revealed that olive oil prevents HeLa cells against oxidative stress by decreasing ROT level, protecting nDNA and supressing the apoptotic effect of oxidative stress.
