Abstract
Bu tez çalışmasında nişasta biyosentezinde görev alan AGPaz enzimi büyük alt ünitesi (LS), küçük altünitesi (SS) ve nişasta dallandırma enzimleri cDNA'larının izolasyonu ve karakterizasyonu yapılmıştır. Aynı zamanda izole edilerek genetik dizileri belirlenen bu genlerin dokuya ve zamana bağlı anlatım profilleri de oluşturulmuştur.Mercimek, baklagiller (Leguminosae) familyasına ait bir bitki olup önemli oranlarda nişasta içermektedir. Türkiye ve gelişmekte olan ülkelerde önemli bir besin kaynağı olan mercimekte nişasta sentezinden sorumlu bu enzimleri kodlayan genler bilinmemektedir.Yapılan çalışma sonucunda tespit edilen yeni gen dizileri NCBI gen bankasında yerverilmiştir. Belirlenen bu gen dizileri bundan sonra bu bitki ile yapılacak olan çalışmalar için temel oluşturacaktır.Tez çalışmasının nihayi hedefi, nişasta sentezinde görev alan bu enzimleri kodlayan genlerin dizilerinin belirlenmesinin ardından ileriki çalışmalarda mercimekte nişasta sentez hızını artıracak çalışmalar yapmak ve bitki veriminin artırılmasını sağlamak olacaktır.Tezin amacı doğrultusunda iklim odalarında büyütülen mercimeklerden RNA izolasyonu ve cDNA sentezi yapılmış, bu cDNA'lardan Polimeraz Zincir Reaksiyonu sonucunda istediğimiz genlere ait parçalar elde edilmiştir. Bu parçalara DNA dizi analizi yapılarak değerlendirilmesi sonucu enzimleri kodlayan genlerin tam dizisi elde edilmiştir.Sonuç olarak izole edilen genlere NCBI gen bankasında, ADP-Glikoz pirofosforilaz büyük altünitesi L1 izoformu ulaşım kodu: GQ861440; ADP-Glikoz pirofosforilaz küçük altünitesi S1 izoformu ulaşım kodu: GQ919066 ile ulaşılabilmektedir. Genetik dizileri bu tez çalışmasında belirlenen bu genler ve izoformlarının kök, gövde, yaprak ve tohumda ifade düzeylerinin farklı olduğu tespit edilmiş olup, aynı dokuya ait örneklerde zamana bağlı bir anlatım düzeyinde farklılık da gözlemlenmiştir. Lentil (Lens culinaris Medik.) is one of the most important members of Leguminosae family with high starch content.Starch biosynthesis occurs by the participation of three main enzymes; ADP-Glucose Pyrophosphorylase (AGPase), Starch Synthase (SS) and Starch Branching Enzyme (SBE). ADP-Glucose pyrophosphorylase catalyses the first commited step of starch biosynthesis in higher plants. Although lentil is an important nutritional source for Turkey and many developing countries, the genes encode for these enzymes haven?t been isolated yet.The ultimate aim is to enhance starch synthesis rate,hence, the yeild of the plant. For this purpose, RNA isolation and cDNA synthesis has performed from the lentil grown in the climate chambers and the fragments of the genes of interest has amplified by Polymerase Chain Reaction (PCR) using these cDNAs. After DNA sequencing we have determined the full lenght of these genes which will be reference for future studies.In this thesis, we have isolated and characterized the cDNAs of AGPase large subunit (LS), AGPase small subunit (SS) and starch branching enzyme (SBE). After the isolation of these genes the time depended and tissue spesific expression profiles have been determined. The expression profiles of these genes and isoforms which were determined in this thesis are observed to be different in various tissues like root, stem, leaf and seed. Also in the same tissue sample a time dependent expression difference has been detected.After the studies the isolated gene sequences were submitted to NCBI genebank. These sequences will be a basic reference for future studies about this plant. The genebank ID numbers are LS-L1: GQ861440, SS-S1: GQ919066.
