Abstract
Astragalus chrysochlorus'da Selenyum Birikimine İlişkin Moleküler ÇalışmalarSelenyum kanserden koruyucu etkilerinin yanı sıra insan sağlığında önemli rolleri olan bir elementtir. Selenyumlu bileşiklerin kanserden koruyucu etkilerinin değişkenlik gösterdiği saptanmış olmakla birlikte, metilselenosisteinin (MeSeCys) en etkili bileşiklerden biri olduğu bilinmektedir. Astragalus cinsi bitkilerin yüksek oranda MeSeCys olmak üzere, selenyumlu bileşikleri biriktirdikleri belirlenmiştir. Bu nedenle MeSeCys biriktirilmesi çalışmaları için uygun bir model oluşturmaktadırlar. MeSeCys oluşumundan sorumlu olan selenosistein metiltransferaz (SMT) enzimini şifreleyen gen Astragalus chrysochlorus bitkisinden, veribankasında bulunan diğer SMT genleri ve daha önceki çalışmamızdan elde edilen dizi bilgileri kullanılarak tasarlanan primerler ile PZR ve RACE PZR yöntemleriyle izole edilmiştir. Bu genin, 1.242 bç. uzunluğunda ve 340 amino asit, 40 kDa ağırlığında proteini şifreleyen açık okuma çerçevesine sahip olduğu belirlenmiştir. AcSMT'nin aminoasit dizisinin A. bisulcatus SMT proteinine % 97 oranında benzediği saptanmıştır. Mitokondri ve kloroplast sinyali içermediği belirlenen AcSMT'nin karboksil ucunda, diğer metil transferazlar gibi çinko bağlayan GGCC motifi içerdiği ve bu bölgeye yakın konumlanmış korunmuş sistein bakiyeleri saptanmıştır. AcSMT geni, Escherichia coli'ye aktarılarak bu bakteride anlatımı sağlanmış ve ürünü olan protein Western Blotlama yöntemiyle belirlenmiştir. Ayrıca bu bakterilerden elde edilen hücre ekstrelerinde yapılan enzim aktivitesiyle AcSMT'nin varlığı saptanmış, farklı selenat uygulamaları ile anlatımı araştırılan AcSMT geninin bitkide sürekli anlatım yaptığı bulunmuştur. AcSMT geninin bakteride klonlanarak ürünün anlatımının başarılması, bu enzimin rekombinant DNA teknolojisi ile üretimine temel oluşturacaktır. Molecular Studies on Selenium Accumulation in Astragalus chrysochlorusSelenium (Se) plays an indispensable role in human nutrition and has been implicated to have important health benefits, including being a cancer preventative agent. While different forms of Se vary in their anticarcinogenic efficacy, methylselenocysteine (MeSeCys) has been demonstrated to be one of the most effective chemopreventative compounds. Astragalus sp. is known for its ability to accumulate high levels of Se with the majority of the selenoamino acids in the form of methylselenocysteine. Therefore, they serve as a good model to study the regulation of MeSeCys accumulation in plants. A cDNA encoding selenocysteine methyltransferase (SMT), the key enzyme responsible for MeSeCys formation, was cloned from Astragalus chrysochlorus using specific primers designed from a fragment that we obtained in our previous work and other SMT genes in databases. AcSMT isolated by PCR and RACE-PCR reactions. This gene is a 1.242 bp cDNA with an open reading frame predicted to encode a 340 amino acid, 40 kD protein. The predicted amino acid sequences of AcSMT shows 97% identity with A. bisulcatus selenocysteine methyltransferase (AbSMT). Analyses of AcSMT showed that it lacks obvious chloroplast or mitochondrial targeting sequences and contains a consensus sequence of GGCC for a possible zinc-binding motif near the C-terminal and a conserved Cys residue upstream of the zinc-binding motif as other related methyltransferases. This cDNA, designated as AcSMT, was functionally expressed in Escherichia coli, and its identity was confirmed by sequence analysis, western blotting and enzyme activity. Expression of AcSMT correlated with the presence of SMT enzyme activity in cell extracts. Examination of AcSMT gene expression in response to selenate treatments showed that the AcSMT transcript was constitutively expressed in plants. We have achieved to clone and express AcSMT in bacteria. This results provide an avenue for producing AcSMT by using recombinant DNA technologies.
