Gökkuşağı alabalığında (Oncorhynchus mykiss) germ hücre izolasyonu üzerine bir çalışma

Abstract
Bu çalışma, ülkemiz iç sularında yetiştiriciliği en fazla yapılan tür olan gökkuşağı alabalığının (Oncorhynchus mykiss) testis dokusundan percoll gradient santrifüj yöntemi ile erken evre germ hücre izolasyonunun gerçekleştirilmesi amacıyla yapılmıştır. Çalışmada İstanbul Üniversitesi Su Bilimleri Fakültesi Sapanca İçsu Ürünleri Üretimi Araştırma ve Uygulama Biriminden temin edilen ortalama 24,76±1,34 cm boy uzunluğunda ve ortalama 171,9±29,10 gram ağırlığındaki 15 adet erkek gökkuşağı alabalığı kullanılmıştır. Balıklara ait testis dokusu enzimatik ayrıştırma amacıyla fosfat tamponlu tuz çözeltisi (PBS) içerisinde % 0.5 trypsin kullanılarak 2 saat süre ile inkübasyona tabi tutulmuştur. İnkübasyın sonrası örnekler 50 µm'lik filtreden geçirilmiş ve 40 µg/ml Deoxyribonuclease I (DNaz I) ve %1 Bovin Serum Albumin (BSA) eklenmiştir. DNaz, ölü hücrelerden Deoksiribo Nükleik Asitin (DNA) serbest bırakılmasından dolayı ortaya çıkan hücre süspansiyonunun pıhtılaşmasını engellemek ve BSA enzim aktivitesinin durdurulması amacıyla eklenmiştir. Filtrasyon işleminden sonra üç farklı percoll gradient grubu (%45 - %10, %50 - %10 ve %55- %10) oluşturulmuştur. Bu işlem sonrası oluşan herbir katmandaki örnek Olympus CX41 mikroskopta incelenmiştir. Bu inceleme sonucunda santrifüj sonrası oluşan en üst katmanda; debris ve spermatidler, orta katmanda erken evre germ hücreleri ve az miktarda spermatidler, dip katmanda yer alan pelette ise eritrosit ve yoğun olarak spermatidler tespit edilmiştir. Orta katmanda tespit edilen erken evre germ hücreleri DEAD (Asp-Glu-Ala-Asp) box Polypeptid 4 Primer Antikoru (DDX4), anti-tavşan IgG- keçide üretilen Floresin İzotiyosiyanat Sekonder Antikoru (FITC) ve 4,6-Diamidino-2-Phenylindole (DAPI) ile immunofloresan işaretlenmesi sonucunda yalnızca DAPI ile boyanan hücrelere ait görüntüler tespit edilmiştir. DAPI tüm testiküler hücrelere ait nukleusları boyama özelliğine sahip olduğundan hücreler arasında spesifik bir ayrım elde edilememiştir. Testis dokusundan alınan histolojik kesitlerin Hematoksilen-Eosin ile boyanması sonucunda, spermatogonya Tip A ve B ve spermatid hücreleri tespit edilmiştir. Doku kesitlerinin floresan boyanması sonucunda ise primer antikor (DDX4) ve sekonder antikor (FITC) ile işaretlenmiş erken evre germ hücreleri tespit edilmiştir. This study was carried out to perform early stage germ cell isolation by percoll gradient centrifugation from the testis tissue of the rainbow trout (Oncorhynchus mykiss) which is the most cultivated species in the inland waters of our country.A total of 15 male rainbow trout with an average length of 24,76 ± 1,34 cm and an average weight of 171,9 ± 29,10 grams obtained from the Sapanca Inland Water Products Research and Application Unit of Faculty of Aquatic Sciences in Istanbul University were used in the study.Testicular tissue of fishes was incubated with 0.5% trypsin in phosphate buffered saline (PBS) for 2 hours for enzymatic separation. After incubation, samples were passed through a 50 μm filter and 40 μg / ml Deoxyribonuclease I (DNase I) and 1% Bovine Serum Albumin (BSA) were added. DNase was added to prevent clotting of the cell suspension resulting from the release of Deoxyribose Nucleic Acid (DNA) from dead cells and BSA was added to stop the enzyme activity. After filtration, three different percoll gradients (45% - 10%, 50% - 10% and 55% - 10%) were formed. The sample on each layer after this procedure was examined under the Olympus CX41 microscope. As a result of this examination, the top layer formed after centrifugation; lipids, debris and spermatids, early-stage germ cells in the middle layer, and erythrocytes and spermatids in the bottom layer of the pellets. The early stage germ cells detected in the middle layer were stained with primer antibody DEAD (Asp-Glu-Ala-Asp) box Polypeptid 4 (DDX4), secondary antibody anti-rabbit IgG (whole molecule) FITC antibody produced in goat, and 4,6-Diamidino-2-Phenylindole (DAPI). Immunofluorescence staining showed only staining with DAPI. Since DAPI has staining properties for all testicular cell nuclei, no specific discrimination between cells has been obtained. Hematoxylin-eosin staining of histological sections from testicular tissue revealed spermatogonia types A and B and spermatid cells. Fluorescent staining of tissue sections revealed early stage germ cells labeled with the primary antibody (DDX4) and the secondary antibody (FITC).
