Abstract
N,N-dimetiltriptamin (DMT), vücutta periferik dokularda sentezlenen bir halusinojendir. Sigma-1 reseptörü (Sig-1R) agonisti olduğu keşfedilen DMT'nin, psikoaktif etkileri üzerinde durulmasına rağmen merkezi sinir sistemindeki fizyolojik rolü halen bilinmemektedir. Sig-1R, aksonal ve dendritik yapıların (neurite) oluşumu, plastisite, ER'dan mitokondriye Ca+2 akışı ve canlılık süreçlerinde önemli rol oynamaktadır. Çalışmamızın amacı, Sig-1R endojen agonisti DMT'nin neurite oluşumu ve nöronal canlılık üzerindeki olası fizyolojik etkilerini araştırmaktır. Bu amaçla, yeni doğan fare hipokampal nöron kültürü yapıldı. Deney gruplarına sırasıyla DMT veya Sig-1R sentetik agonisti SA4503, DMT ve SA4503 öncesinde Sig-1R antagonisti veya inositol 1-4-5-trifosfat reseptörü (IP3R) antagonisti uygulandı. Ardından immünositokimya ile neurite yapıları işaretlenerek konfokal mikroskop görüntülemeleri üzerinden neurite analizi yapıldı. İnsan nöroblastoma hücreleri de sırasıyla DMT veya SA4503 ile ayrıca DMT ve SA4503 öncesinde Sig-1R antagonisti ile muamele edildi, ardından hücrelerde hidrojen peroksit toksisitesi oluşturuldu. Laktat dehidrogenaz testi ile hücre canlılığı, total oksidan kapasite testi ile oksidan parametreler ölçüldü. Sig-1R'nün hücrelerdeki varlığı immünositokimya ve western blot teknikleriyle gösterildi. Çalışmamızda, DMT'nin hipokampal nöronlarda neurite oluşumunu arttırdığı (p<0,001), Sig-1R ve IP3R antagonisti uygulamaları ile bu etkisinin azaldığı görüldü (p<0,001). Ayrıca DMT'nin, hidrojen peroksit toksisitesini ve total oksidan kapasiteyi düşürerek (p<0,001) canlılığı arttırdığı, Sig-1R antagonistinin DMT kaynaklı antioksidan etkiyi azalttığı gözlendi (p<0,001). Endojen agonist olarak DMT'nin bu etkilerinin sentetik agonist kadar güçlü olduğu saptandı. Çalışmamızdan elde edilen veriler, DMT'nin Sig-1R üzerinden gerçekleşen nöroprotektif ve antioksidan etkilere sahip olduğunu, yine Sig-1R üzerinden neurite oluşumunu uyardığını ve bu etkisinin ER ile mitokondri arasındaki Ca+2 taşınmasının regülasyonu üzerinden olduğunu göstermektedir. N,N-dimethyltryptamine (DMT), is a hallucinogen synthesized in peripheral tissues in the body. The majority of research has been focused on the psychoactive effects of DMT, discovered as an endogenous agonist of sigma-1 receptor (Sig-1R) but its physiological role in the central nervous system is still unknown. Sig-1R plays a vital role in neurite outgrowth, plasticity, Ca+2 transport from ER to mitochondria, viability. The aim of this study is to investigate the physiological effects of DMT in neurite outgrowth and neuronal survival. For this purpose, postnatal mice hippocampal neuron culture was performed. Neurons were treated with DMT or Sig-1R agonist SA4503. Sig-1R antagonist or inositol 1,4,5-triphosphate receptor (IP3R) antagonist were also added to culture before DMT and SA4503. Neurites were labeled immunocytochemically. Neurite analysis was performed on confocal microscope images. Human neuroblastoma cells were also treated with DMT or SA4503. Sig-1R antagonist was applied before DMT and SA4503. Then cells were exposed to hydrogen peroxide (H2O2). To determine cell viability and oxidant status, lactate dehydrogenase and total oxidant capacity tests were used. The presence of Sig-1R in cells was investigated by immunocytochemistry and western blotting. In our study, DMT increased neurite outgrowth (p<0.001), Sig-1R and IP3R antagonist treatment reduced this effect (p<0.001). DMT decreased H2O2 toxicity and oxidant capacity thus increased viability (p<0.001), Sig-1R antagonist treatment reduced this effect (p<0.001). Also, DMT were as potent as synthetic agonist. Our findings showed that DMT has neuroprotective, antioxidant effects through Sig-1R, induces neurite outgrowth through Sig-1R regulating Ca+2 transport between ER and mitochondria.
