dc.description.abstractBu çalışmada, tarımsal faaliyetler sonucu sucul ortamlarda toksik etki gösterdiği belirtilen karbamatlı pestisitlerden karbarilin tatlı sularda yaşayan Capoeta capoeta balıkları üzerine etkilerinin histopatolojik ve biyokimyasal yöntemlerle belirlenmesi amaçlandı.Kars Çayı'ndan yakalanan C. capoeta' lar laboratuara getirilerek 300 L'lik tanklara yerleştirildi ve 7 gün süreyle ortama adaptasyonları sağlandı. Her grupta 10 balık olmak üzere 3 gruba ayrıldı. 1.gruptaki balıklar normal su ortamında(kontrol grubu), 2. gruptaki balıklar 0.05 mg/L karbaril içeren, 3. gruptaki balıklar ise 0.1 mg/L karbaril içeren tanklarda 7 gün süreyle bekletildi. Bu süre sonunda balıklardan histopatolojik incelemeler için solungaç, bağırsak ve böbrek örnekleri alınarak %10'luk formaldehit solüsyonunda tespit edildi. Rutin histolojik yöntemlerle parafin bloklar hazırlandı. 3-5 µ kalınlığında kesitler alınarak hematoksilen ve eosin boyama metoduna göre boyanarak ışık mikroskobunda incelendi. Biyokimyasal analizler için alınan karaciğer örneklerinden ise uygun metotlarla homojenatlar elde edildi. Bu homojenatlar santrifüj edilerek süpernatant kısmı ependorf tüplere alındı ve ticari kit kullanılarak otomatik analizördeaspartat aminotransferaz (AST) ve alanin aminotransferaz (ALT) düzeyleri belirlendi.Karbaril uygulaması ile oluşturulan gruplardan elde edilen solungaç, bağırsak ve böbrek dokularında yapılan histopatolojik incelemelerde dejenerasyon, hemoraji ve mononükleer hücre infiltrasyonları tespit edildi. Yapılan biyokimyasal analizler neticesinde ise karaciğer AST ve ALT düzeylerinin karbaril verilen gruplarda, kontrol grubuna göre önemli oranda artış gösterdiği saptandı. Sonuç olarak karbarilin uygulanan doz ve sürede C. capoeta'da toksik etki meydana getirdiği ve organlarının ciddi anlamda hasar gördüğü sonucuna varıldı.
dc.description.abstractIn this study, it was aimed to determine with histopathologic and biochemistry methods the effects of Capoeta capoeta fish living in fresh water with carbaryl, known as a variety of pesticides with carbamate which is reported to show toxic effect in aquatic media as a result of agricultural activity. C. Capotea which were caught in Kars river were brought to the laboratory and placed in tanks with 300 L, and they were adapted to the laboratory environment for seven days. They were divided into three groups, each one consists of ten fish. The fish in the first group (control group) were kept in tap water, the fish in the second group were kept in the tanks with 0.05 carbaryl, the fish in the third group were kept in the tanks with 0.1 mg/L carbaryl for seven days. At the end of the exposure time, gill, intestine and kidney samples were extracted from the fish and fixed into 10 % formaldehit solution for histopathologic examination. Paraffin blocks were prepared with routin histologic methods. Paraffin block were cut 3 – 5 µ thicks and stained in compliance with hematoxylin eosin staining methods and examined in the light microscope. Homogenates were obtained from liver samples with appropriate methods for biochemical analysis. These homogenates were centrifuged and supernatant part was put into eppendorf tubes, and the levels of aspartate aminotransferase (AST) and alanine aminotransaminase (ALT) were determined by using commercial kits. Degeneration, haemorrhage and mononuclear cell infiltration were determined in histopathologic examination of the gill, intestine, liver and kidney tissues obtained from the groups with carbadryl application. As a result of the biochemical analysis it was found that there was more increase in the levels of liver AST and ALT than the control groups. As a result, it was evaluated that the Carbaryl aplication dose and duration were made up toxic effects for C. capoeta and were damaged to internal organs.en_US
