dc.description.abstractBu çalışmada Elazığ ili Baskil ilçesinden toplanan Kickxia lanigera (KL) ve Kickxia spuria (KS) bitki ekstrelerinin fitokimyasal analizi yapıldı ve ekstrelerin biyolojik aktivitesi incelendi. Aktivitesi yüksek olan CHCl3 ve EA ekstrelerinden sekonder metabolitler saflaştırılarak NMR, TOF/MS gibi tekniklerle yapıları aydınlatıldı. GC-MS analizleri sonucunda hekzan ve CHCl3 ekstrelerindeki yağ asidi oranının % 62.5 ve 93.73, hidrokarbon oranının ise % 26.5 ve % 1.86 olduğu belirlendi. HPLC-TOF/MS analizleri sonucunda CHCl3 ve BuOH ekstresinin standart fenolikler açısından fakir olduğu, EA ekstresindeki ana bileşenlerin (mg/kg bitki cinsinden) miktarları ise sinamik asit (5.92), hesperidin (4.7), apigetrin (4.5) ve p-kumarik asit (2.8) olduğu tespit edildi. Antikanser test sonucunda KL bitkisinin CHCl3 ekstresinin, hekzan, EA ve BuOH ekstrelerinden ve 5-FU'den aktif olduğu belirlendi. Antioksidan test sonucunda H2O2 giderme aktivite testinde KS bitkisinin sap kısmının en aktif olduğu (A0.5=27.18 μg/mL) tespit edildi. Kromatografik işlemler ile KL bitkisinin CHCl3 ekstresinden bir tanesi yeni 8 flavonoid (Mosloflavon, Baicalein 5,6,7-trimethyl ether, Salvigenin, Chrysin, Acacetin, Jaceosidin, Hispidulin) ve 1 steroid (Daucosterol) olmak üzere 9, EA ekstresinden ise 2 (Luteolin ve Apigenin) flavonoid olmak üzere toplam 11 bileşik saflaştırıldı. Saflaştırılan bileşiklerin antioksidan aktiviteleri incelendi ve indirgeme gücü aktivitesinde Mosloflavon'un (IC50=82.78±0.05), H2O2 giderme aktivitesinde Chrysin'in (IC50=24.17±2.00) ve yeni bileşiğin (IC50=26.90±2.56) standartlardan yüksek aktiviteye sahip olduğu tespit edildi.
dc.description.abstractIn this study, phytochemical analysis of Kickxia lanigera (KL) and Kickxia spuria (KS) plant extracts collected from Elazığ Baskil carried out and biological activity of the plant extracts were examined. Secondary metabolites were purified from CHCl3 and EA extracts with high activity and their structures were elucidated by techniques such as NMR, TOF / MS. As a result of GC-MS analysis, it was determined that the fatty acid ratio in hexane and CHCl3 extracts was 62.5% and 93.73%, and the hydrocarbon ratio was 26.5% and 1.86%. As a result of HPLC-TOF / MS analyzes, CHCl3 and BuOH extract were poor in terms of standard phenolics, and the amount of main components (in mg/kg plant) in EA extract are cinnamic acid (5.92), hesperidin (4.7), apigetrin (4.5) and p-coumaric acid (2.8). In anticancer test, it was determined that the CHCl3 extract of the KL plant was more active than hexane, EA and BuOH extracts and 5-FU. In the H2O2 removal activity test, the stem part of the KS plant was found to be the most active (A0.5 = 27.18 μg / mL) as a result of the antioxidant test. With chromatographic methods 8 flavonoids (Mosloflavone, Baicalein 5,6,7-trimethyl ether, Salvigenin, Chrysin, Acacetin, Jaceosidin, Hispidulin and New Compound) and 1 steroid (Daucosterol) were purified from CHCl3 extract and 2 flavonoids (Luteolin and Apigenin) from EA extract of KL plant. The antioxidant activities of the purified compounds were examined and it was determined that Mosloflavone (IC50 = 82.78 ± 0.05) in the reduction power activity, Chrysin (IC50 = 24.17 ± 2.00) and New Compound (IC50 = 26.90 ± 2.56) in the H2O2 scavenging activity had higher activity than the standards.en_US
