dc.description.abstract58 ÖZET Köpek ve Kedi Gaitalarından C.difficile İzolasyonu ve Toksin A Saptanması Bu çalışmada evcil hayvanlardan C.difficile izolasyonu ve toksin A'nın saptanması ülkemizde ilk defa yapıldı. C.difficile'nin izolasyonu için 198 köpek (80'i enteritisli, 118'i enteritissiz) ve 68 kedi (26'sı enteritisli, 42'si enteritissiz) gaita örneği Cycloserine-Cefoxitene Fruktoze Agar (CCFA) (Oxoid) besi yerine ekildi. İzole edilen bakterilerin kesin identifikasyonu Analytical Profile Index (API-20A) (BIO MERIEUX) ile yapıldı. Toksin-A saptanması Enzym Linked Fluorescent Assay (ELFA) yöntemiyle Vıtek Immuno Diagnostik Assay Clostridium difficile toxin-A (Vidas CDA.) (BIO MERIEUX) ile yapıldı Köpeklerden toplanan dışkı örneklerinin (80'i enteritisli, 118'i enteritissiz) enteritisli olanların 33'ünden (% 41 ) ve enteritissizlerin 34'ünden (% 29) C.difficile izole edildi. Enteritislilerin 22'sinden (% 28) ve enteritissizlerin 24'ünden (% 20) toksin A antijeni saptandı. 198 köpek gaita örneğinden toplam 67 tanesinden (% 34) C.difficile izole edildi, 46 tanesinden (% 23) ise toksin-A antijeni saptandı. Benzer olarak kedilerden alınan dışkı örneklerinin (26'sı enteritisli, 42'si enteritissiz) enteritisli olanların 9'undan (% 35) ve enteritissiz olanların 12'sinden (% 29) C.difficile izole edildi. Yine enteritisli kedilerin 6'sından (% 23) ve enteritissizlerin 12'sinden (% 29) toksin A saptandı. Çalışılan toplam 68 kedi dışkı örneğinin 21 'inden (% 31) C.difficile izole edildi, 18'inden (% 27) toksin-A antijeni saptandı. dc.description.abstract59 SUMMARY Isolation of Clostridium difficile From Dog and Cat Faeceses and Detection of Toxin A In this study, isolation of C.difficile from domestic animals and detection of toxin A were managed for the first time in our country. Faecal samples of total of 198 dogs, composed of 80 with and 118 without enteritis, and 68 cats, composed of 26 with and 42 without enteritis, were inoculated on Cycloserine-Cefoxitine Fructose Agar (CCFA) for the isolation of C.difficile. The isolates were identified with the aid of Analytical Profile Index (API-20A) (BIO MERIEUX). The detection of toxin-A was carried out by Vitek Immuno Diagnostic Assay Clostridium difficile toxin-A (Vidas CDA) (BIO MERIEUX) test which works according to the Enzyme Linked Fluorescent Assay (ELF A). Among the faecal samples obtained from dogs with (80 individuals) and without (118 individuals) enteritis, C.difficile was isolated from 33 (41 %) and 34 (29 %) samples, and toxin A antigen was detected in 22 (28 %) and 24 (20 %) samples, respectively. Totally, among 198 dog faecal samples tested, C.difficile was isolated from 67 (34 %) and toxin A antigen was detected in 46 (23 %) samples. Similarly, among the faecal samples optained from cats with (26 individuals) and without (42 individuals) enteritis, C.difficile was isolated from 9 (35 %) and 12 (29 %) samples, and toxin A was detected in 6 (23 %) and 12 (29 %) samples, respectively. Totally, among 68 cat faecal samples tested, C.difficile was isolated from 21 (31 %) and toxin A antigen was detected in 1 8 (27 %) samples.
