dc.description.abstractVI. ÖZET Benzenin sıçan ve tavşan karaciğer, böbrek ve serum Alanin transa- minaz (ALT), Aspartat transaminaz (AST), Laktat dehidrojenaz (LDH), Alkalen fosfataz (ALP), Katalaz, Süperoksit dismutaz ve Glutatyon-S transferazm l-kloro-2,4-dinitrobenzen (CDNB) ve l,2-dikloro-2,4- nitrobenzen (DCNB) substratlarma karşı olan aktiviteleri üzerindeki etki leri incelenerek kontrol grubu ile mukayese edilmiştir. Bu çalışmada 5 adet erkek sıçan'a ve 4 adet erkek Yeni Zelanda tavşanına (880 mg/kg/gün) dozunda 3 gün süreyle subkütan yolla benzen verilerek sırasıyla aynı cins 5 adet sıçandan ve 4 adet tavşandan oluşan kontrol grubu ile adı geçen enzimler yönünden mukayese edilmiştir. Sonuçlar aşağıda sıralandığı gibidir: 1. İlk dozdan 71 saat sonra kanları alman, kesilen sıçan ve tavşanların karaciğer ve böbrekleri buz içinde homojenize edilmiş ve 18.000 devirde + 4 derecede santrifüje edilerek elde edilen süpernatan (si- tozol fraksiyonu) da aynı gün içinde tüm enzimlerin miktarları tayin edilmiştir. 2. Sıçanlarda hiçbir dokuda hiçbir enzim kontrole göre farklılık göstermemiştir. 3. Sadece tavşanlarda hem CDNB hem de DCNB'ye karşı karaciğer Glutatyon S-Transferaz aktivitesi kontrol grubuna oranla yüksek bulun muştur.
dc.description.abstractVII. SUMMARY The acute effect of benzene on rat and rabbit hepatic and kidney Ala nine aminotransferase, Aspartate aminotransferase, Alkaline phospha tase, Lactat dehydrogenase, Catalase, Superoxide dismutase and Glutathione S-transferase activities toward the substrates l-chloro-2,4- dinitrobenzene (CDNB), l,2-dichloro-4-nitrobenzene (DCNB) were deter mined and compared to those obtained with control group of these spe cies. In this study 5 male rat and 4 New Zeland male rabbit were admines- tered a single subcutaneous 0.880 mg/kg/day dose of benzene for three days prior to sacrifice. The results were compared with those obtained control groups of rat 5 and rabbit 4 respectively. The results were as fol lowing: 1. After 72h of first dose blood was drawn prior to sacrifice and the liver and kidney of animals were homogenized on ice and centrifuged at 18.000 rpm at + 4UC degree. The activity of the enzymes were determined in the same day as supernatant (cytosol fraction) of tissue were obtained. 2. The results indicated that the activity of all enzymes in the every tissue did not showed any differences in the rats from the controls. 3. Only the activity of Glutathione S-Transferase toward DCNB and CDNB were higher in the rabbit liver than those obtained from the liver of control group.en_US
