Süt ve süt ürünlerinde listeria varlığının araştırılması

Abstract

ÖZET Bu çalışmada süt ve süt ürünlerinde Listeria ile kontaminasyon oranını belirlemek amacıyla İstanbul'un Anadolu yakasındaki çeşitli semtlerden toplanan 370 çiğ süt ve 186 sert peynir örneğinde ELISA (Listeria Tek, Organon Teknika Corp.), Henry illüminasyon ve kültür yöntemleri uygulanarak Listeria araştırılmış, izole edilen Listeria suşlannda biyokimyasal yöntemlerle tiplendirme yapılmıştır. Sert peynir örnekleri marketlerden, çiğ süt örnekleri ise çiftlik lerden direkt olarak inek memelerinden toplanmıştır. Çiğ süt örneMerinin 15'inde (%4.1) ELISA ile, 17'sinde (%4.6) ise kültür yönte mi ile Listeria varlığı saptanmıştır. Sert peynir örneklerinin 3'ünde (%1.6) ELISA ile, 5'inde (%2.7) ise kültür yöntemi ile Listeria varlığı sap tanmıştır. 13'ü çiğ süt örneklerinden, 5'i de sert peynir örneklerinden olmak üzere kültür ile izole edilen toplam 22 Listeria susunun 10'i (%45.5) Listeria monositogenes, 7'si (%31.8) Listeria innocua 3'ü (%13.6) Listeria welshimeri, 2'si (%9.6) ise Listeria ivanovi' den oluşmaktaydı. Bizim bulduğumuz oranlar daha önce ülkemizde yapılan çalışmalarda elde edilen oranlara göre daha düşük bulunmuştur. Özellikle sütler açısından bu fark süt örneklerinin alt yapısı ve hijyen koşulları daha iyi durumda olan İstanbul bölgesinden toplanması ve ineklerden direkt olarak alınması ile ilgili olabilir. -69-

SUMMARY In this study, to find the presence of Listeria contamination in milk and milk products, we examined Listeria in 370 raw milk and 186 hard cheese samples which obtained from various areas of Anotolian side of Istanbul, by ELISA(Listeria Tek, Organon Teknika Corp.) and culture methods, typed the isolated Listeria by biochemical methods. Hard cheese samples were obtained from markets and raw milk samples were obtained from farms by getting directly from the breasts of cows. Listeria is found in 15(%4.1) of raw milk samples with ELISA and 17(%4.6) with culture method. And in hard cheese samples Listeria were found in 3(%1.6) with ELISA and 5(%2.7) with culture method. From 17 Listeria strains that is obtained of raw milk samples and 5 that is obtained of hard cheese samples total 22 Listeria strains, 10 (%45.5) were L. monocytogenes, 7(%31.8) L. innocua, 3(%13.6) L. wels- himeri, 2(%9. 1) L. ivanovii. The ratios that we found are lower than the ratios that were stu died in our country before. This difference may be because the milk samples were directly taken from breasts of cows and from Istanbul co untry which the higienic conditions and substructure are better. 70
