dc.description.abstract | Bu tez çalışmasında; Elajik asidin rat kalvaryalarında oluşturulan kritik boyutkemik defektlerinde meydana gelen yeni kemik dokusuna etkisinin histopatolojikolarak değerlendirilmesi amaçlanmıştır.Bu çalışmada toplam 80 adet Wistar cinsi ratın genel anestezisi 8 mg/100gintra-musküler ketamin ile sağlandı ve ratlar randomize 4 gruba ayrıldı.1. Kontrol Grubu: 20 adet rat üzerinde 8 mm kalvaryal kemik defektioluşturuldu, herhangi bir işleme tabi tutulmadan defekt boş bırakıldı ve yara yeriprimer süture edildi. Deneklerden 10 tanesi 4. hafta sonunda, geriye kalan 10 taneside 8. hafta sonunda sakrifiye edildi.2. Deney Grubu: 20 adet ratta 8 mm kalvaryal kemik defekti oluşturularakdefektler boş bırakılarak yara yeri primer süture edildi.Ratlara günlük 50mg/kg olacakşekilde 4 hafta boyunca oral gavaj yöntemiyle sistemik olarak elajik asit verildi.Deneklerden 10 tanesi 4. hafta sonunda, geriye kalan 10 tanesi ise 8. hafta sonundasakrifiye edildi.3. Elde edilensonuçlar Kruskal-Wallis ve Mann-Whitney U testleri kullanılarak değerlendirildi.XFarklı parametreler ele alınarak yaptığımız deneysel çalışmadeğerlendirildiğinde osteogenezisi hızlandırması bakımından elajik asidin grefteüstünlük sağlamadığı düşüncesindeyiz. 4.ve 8.hafta bulguları değerlendirildiğinde yenikemik formasyonu açısında en iyi sonucu sistemik olarak uygulanan elajik asit + greftuygulanan gruptan elde ettik.Anahtar Kelimeler:1. dc.description.abstract | This research aims to stimulate osteogenesis with graft materials placed onbone tissue and use its osteoconductive feature; and to evaluate histopathologicallythe effect of ellagic acid on osteogenesis by increasing osteoblast cells.In this research, general anesthesia of totally 80 Wistar rats were providedwith 8 mg/100g intra-muscular ketamine and the rats were divided into 4 groupsrandomly.1. Control Group: Calvarial bone defect was created on 20 rats, the defectwas left empty without any intervention and the wound area was sutured. 10 of therats were sacrificed at the end of 4th week and the rest 10 rats were sacrificed at theend of 8th week.2. Experimental Group: 8mm- calvarial bone defect was created on 20 rats,the defect was left empty without any intervention and the wound area was sutured.Ellagic Acid was orally administered to rats for 4 weeks, by means of a gastric tube50mg/kg body weight/day. 10 of the rats were sacrificed at the end of 4th week andthe rest 10 rats were sacrificed at the end of 8th week.3. Experimental Group: 8mm-calvarial bone defect was created on 20 rats.Alloplastic bone graft was treated on prepared defect. 10 of the rats were sacrificed atthe end of the 4th week and the rest 10 rats were sacrificed at the end of 8th week.4. Experimental Group: 8mm-calvarial bone defect was created on 20 rats.Alloplastic bone graft was treated on prepared defect. Ellagic Acid was orallyadministered to rats for 4 weeks, by means of a gastric tube 50mg/kg bodyweight/day. 10 of the rats were sacrificed at the end of the 4th week and the rest 10rats were sacrificed at the end of 8th week.XISpecimens obtained at the end of the research were evaluatedhistopathologically at Dicle University, Medicine Faculty, the laboratory ofHistology Department. Obtained results were assessed using Kruskal-Wallis andMann-Whitney U tests.When the research which was conducted considering different parameters isevaluated, it is thought that ellagic acid is not superior to graft in terms of acceleratingosteogenesis. When the results of 4th and 8th weeks are evaluated, the best results areseen in graft+ ellagic acid group in terms of new bone formation.
