dc.description.abstractSUMMARY The Effects of Transection and Right -Left Hemisectiones of Spinal Cord on the Acetylcholinesterase Histochemistry, Catecholamine Histochemistry and Morphology of Adrenal Medulla. It's now accepted that adrenal medulla is innervated by preganglionic cholinergic neurons present in the T7_10 segments of the intermediolateral columna of spinal cord, which is under brain stem and hypothalamus control. There is also a convincing evidence that autonomic pathways raising from brain stem and hypothalamus crosses within the spinal cord. In this study we aimed to determine the course of this otonomic pathways by evaluating the adrenal medulla morphology after performing, right and left hemisections or transsections at the level of T!-T2. We used Acetylcholinesterase histochemistry, Potassium dicromate fixation+Giemsa staining and Hematoxy line-Eos ine staining to ascertain the content and morphological structure of cromaffin cells and preganglionic cholinergic fibres projecting to them. Spraque Dawley rats, anestethized by intraperitoneal Na-penthotal enjection were used. After performing transsections, and right and left hemissection, at the level of Tx-Tj * adrenals removed rapidly and immersed immediately in to the appropriate fixatives, then they mounted on slides and dyed.Asetilkolinesteraz boyama değerlendirilmesi yapılamadı. Potasyum dikromat f iksasyonu+Giemsa boyama ile katekolamin içeriğinin morfolojik görünümlerinin değişmediği görüldü. Hematoksilen-Eosin boyama yönteminden sonra yapxlan kesitlerde medulla'yı oluşturan kromafin hücrelerde sentez artışı olarak değerlendirilen eukromatik nukleus sayısında, stoplazmik materyelde ve basofilide artış gözlendi. Deneyde, sağ ve sol yarı kesilerde morfolojik farklılıklar görülürken, tam keşi grubunda her iki taraf medulla kromafin hücrelerinde bu farklılıklar yoktu, hücrelerin görünümleri benzerdi. Sağ yarı keşi yapılan grupta, sağ taraf medullada tam keşi yapılan gruplardaki morfolojik bulgulara benzer görünüm bulunurken, sol taraf medullada da benzer morfolojik bulgular görüldü. Sol yarı keşi yapılan grupta ise sol taraf medullada tam keşi bulguları varken, sağ taraf medulla bulguları kontrol grubu ile benzerdi. Elde edilen bulgulara dayanılarak medulla spinalis içinde seyreden, adrenal medulla innervasyonunda rol alan, otonom liflerin keşi seviyesi altında çapraz laş tıkları kanısına varıldı. There was no signifieiant difference in the content and morphological structure of cromaffin eels by Potassium dicromate f ixsation+Giemsa staining method. However, by Hematoxyline-Eosine staining method a signifieiant enhance in the stoplasmic material and basophily of the cromaffin cells with an eukromatic nucleus in it was found. Although there was considerable morphological divergence in the adrenal medullas of right and left hemisectioned group, the transsectioned ones were similar. We have also confirmed morphological findings resembling transsections in both adrenal medullas of right hemisectioned group. However, in the left hemisec toned one left medulla indicated less morphological alterations while the right medulla was similar to the control groups. Consequently; it is possible to say that autonomic pathways coursing within the spinal cord and projecting to the adrenal medulla crosses below the level of transection.
