dc.description.abstractÖZET Bu çalışmada sağlıklı tavuklardan (I. grup) 26 ve hastalıklılardan (II. grup) 42 Escherichia coli (E.coli) susu izole edildi ve biyolojik ve antijenik özellikleri incelendi. İnsan `O` grubu, tavuk, kobay ve tavşan eritrositleriyle yapılan aglütinasyon denemelerinde hemaglütinasyon pozitivite oranlarının I. grupta, II. gruptan daha yüksek olduğu dikkati çekti. Salt Agregasyon Testi'nde I. grupta % 61.5, Il.'de % 28.6 hidrofobisite belirlendi. Günlük civcivler için I. grup izolatlann % 46.1'i, II. gruptakilerin % 100'ü patojenik bulundu. Sırasıyla I. ve II. grup izolatlar % 46.1 ve % 33.3 oranında enteropatojen olarak değerlendirildi. Enterotoksijenik yönden ise iki gruptaki izolatlann oranlan % 23.0 ve % 16.6 pozitifti. Tüm enterotoksijenik suşlar aynı zamanda enteropatojenikdl Adonitol'lü Simmon's Citrate Agar' da üreme ve Congo Red. boyasına bağlanma özelliklerinin tüm suşlar için patojenite kriteri olarak ele alınamayacağı ortaya çıktı. Enteroagregatifis.co/flerin her iki grupta oranlan % 15.4 ve % 11.9'du. İncelenen E.coli izolatlarının çoğunun `Ol`, `02 ve `078` somatik antijenlerine sahip olduğu tespit edildi. Serotip ile patojenite arasında bir ilişki görülmedi. Her iki izolat grubunda da test edilen antibiyotiklere (penisilin, ampisilin, gentamisin, tetrasiklin, nitrofurantoin, enrofloksasin, norfloksasin, sulfametaxazol/trimetoprim, kloramfenikol, amoksisilin) karşı çoklu dirençlilik saptandı. Sonuçlar, `Civciv Patojenite Testi'nin` uygulanan testler içinde, patojenik E. co//`leri ortaya koyabilen en güvenilir test olduğunu ve diğer testlerin de izolatlann farklı patojenite özelliklerinin araştınlmasmda yararlı olabileceğini gösterdi. dc.description.abstractSUMMARY Investigation of Biological and Pathogenicity Characteristics of E. coli Strains Isolated from Healthy and Diseased Chickens In this study, 26 Escherichia coli (E.coli) strains from healthy chickens (group I) and 42 from diseased chickens (group II) were isolated and examined for their biological and antigenic characteristics. In the aglutination tests performed with human `O` group, chicken, guinea pig, and rabbit erythrocytes, being higher hemaglutination positivity rates in group I than those in group II attracted the attention. In Salt Aggregation Test, hydrophobicity were determined at ratios of 61.5% in the group I and 28.6% in the group II. For day old chicks 46.1% of E.coli isolates in the first group and 100% in the second group were found to be pathogenic. Enteropathogenic E.coli rates in the first and second groups were 46.1% and 33.3% respectively. Percentages of the isolates of group I and II enterotoxigenicity were determined to be 23.0 and 16.6. All the enterotoxigenic isolates were also enteropathogenic. Results showed that `growth on Simmon's Citrate Agar with Adonitol` and `Congo Red binding` properties would not be considered to be criteria for pathogenicity. Rates of Enteroaggregative E.coli isolates in both groups were 15.4% and 11.9%. Most of the isolates studied were found to have `01`, `02` and `078` somatic antigens. It was seen that pathogenicity did not depend on antigenic types of E.coli isolates. Multiple-antibiotic resistancy were determined in the isolates of both groups against antibiotics (penicillin, ampicillin, gentamicin, tetracycline, nitrofurantoin, enroflaxacin, norfloxacin, sulfa+trim, chloramphenicol, amoxicillin) examined. Results revealed that in order to decide ifan E.coli isolate was pathogenic, only one test `Chick Pathogenicity Test` used in this study, was reliable, and other tests could be useful for checking the other characteristics of the isolates.
