Yerli portakal çeşitlerinin karotenoid yapıları üzerinde araştırmalar

Abstract

11 ÖZ Yerli Portakal Çeşitlerinin Karotenoid Yapılan Üzerinde Araştırmalar Portakal suları veya konsantratlarının saflık kontrolünde karotenoidler ve karotenin toplam karotenoidlere oranı büyük öneme sahiptir. Yerli portakal çeşitlerinde karotenoid maddeler dağılımını belirlemek amacıyla bu çalışma planlanmıştır. Bunun için genelde 1988-1989 ve 1989-1990 ürünü yerli portakal çeşitleri ve karşılaştırma amacıyla ülkemizde üretilen birkaç yabancı çeşit portakal örnekleri ile çalışılmıştır. 20'si iki ayrı yıla ve üç ayrı yetiştirme yöresine ait yabancı çeşit, 31 'i dört ayrı yöreye ait yerli çeşit portakal örnekleri olmak üzere toplam 51 örnek incelenmiş tir. Bu örneklerden 3'ü 1990-1991 yılı ürünü Karadeniz portakallarına aittir. Portakal örneklerinde karotenoid maddeler dağılımından başka meyve ağırlığı, kabuk oranı, posa oranı, meyve suyu verimi yanında genel meyve suyu bileşimi de belirlenmiştir. Meyve suyu analizleri arasında kuru madde, C-vitamini (askorbik asit), titrasyon asitliği, pH, formol sayısı, indirgen şeker, toplam şeker, sakkaroz, fosfor, kükürt, kalsiyum, demir, bakır, mangan, magnezyum, çinko, potasyum ve sodyum tayinleri yer almıştır. örneklerden ekstraksiyonla elde edilen toplam karotenoidler, kolon kromatografisiyle fraksiyonlara ayrılmış ve bu fraksiyonların 450 nm'deki absorbansları okunarak, saf 3-karoten standardına göre konsantrasyonları

Ill hesaplanmış; ayrıca ince tabaka kromatografi yöntemi ile doğrulamaları yapılmıştır. Karoten fraksiyonunun toplam karotenoidlere oranı tüm örneklerde % 6'nın altında belirlenmiş ve kaynaklarda, saf portakal suları için verilen değerlere uygun bulunmuştur. % 20'nin altında belirlenen karoten ester fraksiyonu ile de benzer sonuç saptanmıştır. ABSTRACT A Research On The Carotenoid Distribution of Local Orange Varieties Carotenoids, and the ratio of carotenes to the total carotenoids are very important in controlling the purity of orange juices and orange juice concentrates. The main subject of this reseacrh was determining the carotenoid distribution of local orange varieties. For this reason the local orange varieties and for comparision of the results a few foreign orange varieties grown in our country, generally in 1988-1989 and 1989-1990 production period were obtained. 20 of the total 51 samples were chosen as foreign samples grown in three different distincts and 31 of them as local samples grown in four different distincts in two consecutive years. 3 of the total 51 samples were grown in Black Sea region in 1990-1991 production period. The carotenoid distribution, fruit weight (in grams), ratio of pulp, orange juice yield and also the general fruit juice composition were determined. The orange juice analysis included dry matter, vitamin C (ascorbic acid), titratable acidity, pH, formol number, invert sugar, totalIV sugar, saccharose, phosphorus, sulphur, calcium, iron, copper, manganese, magnesium, zinc, potasium and sodium. The total carotenoids, extracted from the samples were seperated into fractions by column chromatography, the absorbances of each fractions were determined at 450 nm and by the help of pure 3 -carotene standard the concentration of each fraction was calculated. Also the fractions were applied to thin layer chromatography plates and after development, the results of column chromatography were proved. The ratio of carotene fraction to total carotenoids was found below 6 % for all the samples. This ratio is a purity criteria for orange juices and must be below 10 % and according to some research workers even below 6 %.Also the ratio of carotene ester fraction must be below 20 % in a pure orange juice. In this research this ratio was found below 20 %.
