dc.description.abstractScrophulariaceae familyasına ait olan Verbascum antiochium Boiss. türünün in vitro antimikrobiyal ve antioksidan aktivitesi belirlenmiştir. Artan polarite ve direkt metanol özütleme yöntemleriyle elde edilen polar ve non-polar özütlerin antimikrobiyal aktivitesi kuyu difüzyon yöntemi ile dokuz Gram-pozitif, altı Gram-negatif bakteri ve bir fungus üzerinde çalışılmıştır. Non-polar hekzan ve diklorometan özütlerinde belirgin bir antibakteriyel etki gözlenmezken, metanol/kloroform ve polar metanol/su özütlerinde artan bir etki gözlenmiştir. En yüksek antibakteriyel etki direkt metanol özütünde saptanmıştır. Fungus üzerinde sadece hekzan özütünün antifungal aktivite gösterdiği belirlenmiştir.Antioksidan kapasite direkt metanol özütü kullanılarak DPPH (2,2-difenilpikrilhidrazil) ve beta-karoten renk açılımı yöntemleriyle test edilmiştir. Özüt ve pozitif kontrol olarak kullanılan BHT ve askorbik asidin, DPPH radikalinin %50 inhibisyon sağlayan IC50 değerleri sırasıyla 4.8 mg/ml, 1.53 mg/ml ve 0.46 mg/ml olarak bulunmuştur. Beta-karoten renk açılım testinde, 48. saatin sonunda V. antiochium bitki özütünün linoleik asit oksidasyonunu % 88.58, BHT' nin ise % 92.47 oranında inhibe ettiği belirlenmiştir.
dc.description.abstractThe antimicrobial and antioxidant activities of Verbascum antiochium Boiss. species, a member of Scrophulariaceae family, were determined. The polar and non-polar extracts obtained by increasing polarity and direct methanol extraction were tested for antimicrobial activity by using well diffusion methods against nine gram positive bacteria, six gram negative bacteria and one fungus. While an increasing antibacterial effect was observed on nonpolar methanol-chloroform and polar methanol/water extracts, non-polar parts including hexane and dichloromethane extracts exhibited weak antibacterial effect. The highest antibacterial activity was determined on direct methanolic extract. Among the extracts, only hexane extract showed antifungal activity.The antioxidant capacities of methanolic extract of V. antiochium was determined with DPPH (2,2-diphenylpicrylhydrazil) and beta-carotene bleaching test system. The 50% (IC50) inhibition of methanolic extract, and BHT and ascorbic acid, used as positive controls, on the free radical DPPH were calculated according to results of DPPH method. IC50 of extract, BHT and ascorbic acid were determined as 4.8 mg/ml, 1.53 mg/ml and 0.46 mg/ml, respectively. The % inhibition of linoleic acid oxidation taken on depending on time was evaluated according to blank. At the end of the forty-eight hours, while the methanolic extract of V. antiochium exhibited % 88.58 inhibition on the oxidation of linoleic acid, BHT, positive control, showed % 92.47 inhibition.en_US
