Metilfenidat`ın kötüye kullanımında idrar ve tükrükte belirlenmesi

Abstract

Son yıllarda metilfenidat üretimi artmış bulunmaktadır ve bunun % 90'ı A.B.D. de tüketilmektedir. İlacın elde edilebilirliğinin artması kötüye kullanımı korkusunu da arttırmaktadır; daha çok gençler Ritalin'i ezerek burundan üflemekte ve reçete edilen ilacın tek tableti 5 $'a satılmaktadır. Son yıllarda acil servise gelen metilfenidat'ın kötüye kullanımı ile ilgili olgular da gözönüne alındığında, dikkatleri bu konuya çevirebilmek için ?metilfenidat'ın kötüye kullanımında idrar ve tükrükte belirlenmesi? konulu çalışma yapılmıştır.Bu nedenle metilfenidatın analizi için yeni, basit, duyarlı, doğru ve kesin yüksek performanslı ince tabaka kromatografik metodu geliştirildi ve valide edildi. Optimizasyon çalışmaları İstanbul Üniversitesi Adli Tıp Enstitüsü Toksikoloji Laboratuarında 5 basamakta gerçekleştirildi. Bu çalışmada validasyon çalışmaları için incelenmekte olan parametreler : yürütme oda sıcaklığında 2 bölmeli cam kabin içinde, mobil faz olarak Diklorametan:Aseton:Amonyak ( 7:3:0.2) (v/v) kullanılarak gerçekleştirildi. Metilfenidat için Rf değeri (Rf 0.52 ± 0.02) olarak elde edildi. Tarama 200 nm absorbansta gerçekleştirildi. Kalibrasyon eğrisinin doğrusal regrasyon analizi 100?1000 ng/ ml konsatrasyon aralığında pik alanları arasındaki iyi doğrusallık ilişkisi (r2 = 0.99931) olarak gösterildi. Metod kesinlik,geri kazanım, spesifiklik, tanımlama ve ölçüm limitleri için valide edildi. Tanımlama ve ölçüm limitleri sırasıyla 75 ve 225 ng/spot'dir. İdrar ve tükrükteki geri kazanım % 75 ve % 93 bulundu.

At last recent years the production of ritalin is increased and 90 % of these are comsumed in U.S.A. The easy purchase of the drug increase the fear of abuse mostly the young people crush ritalin snorting and the one tablet of prescribed drug is sold for 5 dollars lastly. At the last recent years the cases consulted of abuse of ritalin to the emergency room in Turkey is taken into account for call attention to this issue the subject investigation of methylphenidate abuse in urine and saliva is done as thesis .Therefore a new, simple, sensitive, accurate and precise high-performance thin-layer chromatographic method for quantification of methylphenidate was developed and validated. Optimization studies take form 5 steps at Toxicology Laboratory of Forensic Science Institute, İstanbul University. In this study, the following parameters were taken under consideration for validation studies :linear ascending development with Diklorametan:Aseton:Amonyak ( 7:3:0.2) (v/v) as the mobile phase was performed at room temperature (25 ± 2°C) in a twin-trough glass chamber. Compact bands (Rf 0.52 ± 0.02) were obtained for methylphenidate . Scanning was performed in absorbance mode at 200 nm. Linear regression analysis of the calibration plots showed good linear relationship between peak area (r2 = 0.99931) in the concentration range 100?1000 ng/ spot. The method was validated for precision, recovery, robustness, specificity, and detection and quantification limits. The limits of detection and quantification were 75 and 225 ng/spot ,respectively. The average urine and saliva recovery of the method was 75 % and 93 %.
