dc.description.abstractBu çalısmada,bazı bitki gelisim düzenleyicilerinin (BGD), paraoksonaz(PON1) ve karbonik anhidraz izoenzimler (hCA-I ve hCA-II) üzerinde in vitroetkileri arastırılmıstır. Bu amaçla kullanılan PON1, Sepharose-4B-L-tirozin-1-naftilamin kimyasal yapısına sahip jel kullanılarak insan serumundan saflastırılmıstır.Arastırmamızda kullanılan hCA-I ve hCA-II izoenzimleri ise Sepharose-4B-Ltirozin-p-aminobenzosülfonamit kimyasal yapısına sahip afinite kolonu kullanılarakinsan eritrositlerinden saflastırılmıstır. ?zoenzimlerin saflığı, Sodyum Dodesil SülfatPoliakrilamit Jel Elektroforezi ile (SDS-PAGE) kontrol edilmistir.BGD olarak tarımda yaygın olarak kullanılan, indol-3-asetik asit (IAA),indol-3- bütirik asit (IBA), giberelik asit (GA3) ve kinetin (Kn) adlı bilesiklerseçilmistir. Kn hariç arastırmamızda kullanılan BGD bilesiklerinin tamamı PON1enzimini önemli ölçüde inhibe ettiği saptanmıstır (IC50= 63,87- 89,48 ?M). IAA veIBA bilesikleri, hCA-I ve hCA-II izoenzimlerini inhibe ederken (IC50= 75,47- 78,13?M), GA3'in her iki enzimi aktive ettiği tespit edilmistir. Ayrıca Kn, hCA-IIizoenzimini aktive ederken hCA-I izoenzimini inhibe ettiği saptanmıstır (IC50= 62,88?M).AAHTAR SÖZCÜKLER: ?ndol-3- asetik asit, indol-3-bütirik asit,giberelik asit, kinetin, paraoksonaz, karbonik anhidraz, in vitro.
dc.description.abstractIn this study, the effect of some plant growth regulators (PGR) wereinvestigated on paraoxonase (PON1) and hCAI and hCAII ezymes in vitro. For invitro inhibition studies, firstly, paraoxonase (PON1) was purified from human serumusing Sepharose-4B-L-tyrosine-1-napthylamine hydrophobic gel. Secondly, humanhCA-I and hCA-II from human erythrocyte were purified with Sepharose-4B-Ltyrosine-p-aminobenzosulfonamide affinity chromatography. The purity ofisoenzymes were checked by SDS polyacrilamid gel electrophoresis.Indole-3-acetic acid (IAA), indole-3-butyric acid (IBA), gibberellic acid(GA3) and kinetin (Kn), which were widely used in agriculture, were chosen as plantgrowth regulators. From the inhibition studies, it was determined that apart from Kn,all plant growth regulators used in this study inhibit PON1 enzyme activity insignificant level (IC50= 63,87- 89,48 ?M). hCA-I and hCA-II isoenzymes activitywere inhibited by IAA and IBA (IC50= 75,47- 78,13 ?M). On the contatry, GA3activates both enzymes. In addition it was determined that Kn activates hCA-II butleads to inhibiton effect on hCA-I (IC50= 62,88 ?M).KEY WORDS: Indole-3-acetic acid , indole-3-butyric acid , gibberellic acid,kinetin, paraoxonase, carbonic anhydrase, in vitro.en_US
