dc.description.abstractETA reseptör antagonisti BQ-123'ün STZ ile diyabet oluşturulmuş sıçalarda leptin düzeylerine olan etkileri araştırılarak diyabette iki önemli endojen faktör, leptin ve ET-1 arasındaki ilişki değerlendirildi.Çalışmada, 24 adet Wistar Albino erkek sıçan üç gruba ayrıldı; Kontrol, STZ, STZ+BQ-123 grupları. Deneysel diyabet, tek doz 60 mg/kg STZ i.p. uygulanarak oluşturuldu. STZ+BQ-123 grubundaki sıçanlara 39. ve 40. günlerde olmak üzere (2 mg/kg + 2 mg/kg) toplam 4 mg/kg BQ-123 i.v. verildi. Son BQ-123 uygulamasından 6 saat sonra alınan plazmada PC (Protein Karbonil), TBARS (Tiyobarbitürik Asit Reaktif Maddeleri), NO (Nitrik Oksit), leptin, Na+, Cl-, K+ düzeyleri araştırıldı.Deney sonunda, STZ (p=0.0001) ve STZ+BQ-123 (p=0.015) grubundaki sıçanların ağırlıkları, Kontrol grubuna göre anlamlı olarak düşüktü. Kan glikoz düzeyleri STZ ve STZ+BQ-123 grubu sıçanlarda Kontrol grubuna göre anlamlı derecede yüksekti (p=0.0001). Plazma leptin düzeylerinde gruplar arasında anlamlı bir fark bulunamadı (p=0.055). Plazma TBARS düzeyi, STZ grubunda, Kontrol (p=0.0001) ve STZ+BQ-123 (p=0.0004) grubuna göre anlamlı olarak yüksekti. PC düzeylerinde gruplar arasında istatistiksel bir fark bulunamadı (p= 0.665). NO düzeyleri STZ (p=0.001) ve STZ+BQ-123 (p=0.0001) grubunda Kontrol grubuna göre anlamlı olarak düşüktü. Serum Na+ konsantrasyon değerleri, STZ grubunda Kontrol grubuna göre anlamlı derecede düşüktü (p=0.001). Serum K+ konsantrasyonu değerleri arasında anlamlı fark yoktu (p= 0.375). Serum Cl- konsantrasyonu değerleri, STZ grubu ve (p=0.0001) ve STZ+BQ-123 (p=0.0001) grubunda Kontrol grubuna göre anlamlı olarak düşüktü.Sonuç olarak, STZ ile diyabet oluşturulmuş sıçanlarda Leptin, PC, K+ düzeylerinde bir değişiklik meydana gelmediği; NO, Na+ ve Cl- düzeylerinde azalma ve TBARS düzeylerinde ise artış bulundu. dc.description.abstractWe aimed to evaluate the relationship between leptin and ET-1, both of which are known as important endogenous factors in diabetes, by investigating the influence of BQ-123, an ETA receptor antagonist, on leptin levels in rats with diabetes induced by STZ.In this study, 24 male Wistar-albino rats were divided into three groups; Control, STZ, STZ+BQ-123 groups. Experimental diabetes was induced by delivering a single dose of 60 mg/kg intravenous STZ. The rats in the STZ+BQ-123 group received 2 mg/kg i.v. BQ-123 at 39 and 40 days (4 mg/kg in total). The plasma specimens collected 6 hours after the last BQ-123 delivery were studied for PC (Protein Carbonyl), TBARS (Thiobarbituric Acid Reactive Substances), NO (Nitric Oxide), leptin, Na+, Cl-, and K+ levels.At the end of the experiment, the weights of rats in the STZ (p=0.0001) and STZ+BQ-123 (p=0.015) groups were significantly lower compared with the values in the Control group. Blood glucose levels were significantly higher in the STZ and STZ+BQ-123 groups than in the Control group (p=0.0001). There was no significant difference between the groups with regard to leptin levels (p=0.055). Plasma TBARS level was significantly higher in the STZ group than in the Control (p=0.0001) and STZ+BQ-123 (p=0.0004) groups. No statistically significant difference was observed between the groups in terms of PC concentration. NO levels were significantly lower in the STZ (p=0.001) and STZ+BQ-123 groups than in the Control group (p=0.0001). Serum Na+ concentration was significantly lower in the STZ group than in the Control group (p=0.001), whereas there was no significant difference between the STZ+BQ-123 and Control groups. No significant difference was observed between the groups relative to K+ values. Cl- values were significantly lower in the STZ (p=0.0001) and STZ+BQ-123 (p=0.0001) groups than in the Control group.In conclusion, while rats with STZ-induced diabetes demonstrated no changes with regard to Leptin, PC and K+ levels, they exhibited reduced NO, Na+ and Cl- concentrations and significantly elevated TBARS levels. BQ-123 was found to have no significantly impact over the Leptin levels and decreasing NO, Na+ ve Cl- concentrations in diabetes, whereas it was observed to have a positive effect by reducing increasing TBARS levels in diabetic rats.Key Words: Diabetes, Leptin, BQ-123, Endothelin-1, Streptozotocinen_US
