dc.description.abstractBu çalışma dünya üzerinde 161, Türkiye florasında 47 türle temsil edilen Tanacetum cinsine ait Tanacetum alyssifolium (Bornm.) Grierson (İliç papatyası) bitkisinde mevcut olan sekonder metabolitlerin kromatografik yöntemler ile izole edilmesi, spektroskopik metotlarla yapılarının belirlenmesi ve antiproliferatif ve antioksidan aktivitelerinin incelelenmesi amacı ile gerçekleştirildi. Bu amaçla, bitkinin toprak üstü kısmı etil asetat, n-bütanol ve su fazı olmak üzere üç çözücü sisteminde ekstrakte edildi. N-bütanol ekstraktı kolon kromatografisi işlemlerine tabi tutularak iki farklı bileşik izole edildi. İzole edilen bileşiklerin yapısı spektroskopik metotlarla (1D-NMR, 2D-NMR, HPLC-TOF) aydınlatılarak yapılarının Luteolin-7-O-β-D-glikozit ve Fraxidin olduğu belirlendi. İzole edilen bileşiklerin antioksidan aktiviteleri, DPPH, ABTS serbest radikallerini giderme ve metal iyonlarını şelatlama kapasitelerine göre değerlendirildi. Antiproliferatif aktiviteleri ise HeLa (İnsan Rahim Kanseri) ve C6 (Sıçan Beyin Tümörü) hücre hatları üzerinde anti-kanser ilaç olarak kullanılan 5-Florourasil (5-FU) ile dört farklı dozda (5, 25, 50, 100 µg/mL) karşılaştırılarak incelendi.
dc.description.abstractThis study was carried out with the aim of isolation of secondary metabolites present in Tanacetum alyssifolium (Bornm.) Grierson plant belonging to Tanacetum genus, which is represented by 161 species in the world and 47 species in Turkey, by means of chromatographic methods and determination of structures by spectroscopic methods as well as determination of their antioxidant and antiproliferative activities.For this purpose, Tanacetum alyssifolium plants was collected from the foothills of Munzur Mountains in Erzincan province and dried at room temperature. The upper part of ground plant was extracted with ethyl acetate/ n-butanol and water solvent system. The n-butanol extract was subjected to the procedure of column chromatography. The structures of the isolated compounds were elucidated by spectroscopic methods (1 H-NMR, 2 D-NMR, HPLC-TOF) to reveal that Luteolin-7-O-β-D-glikozit and fraxidine. The antioxidant activities of the isolated compounds were evaluated according to the DPPH and ABTS free radical scavenging and metal ion chelating capacities. Antiproliferative activities of the samples was investigated on HeLa (Human Rectum Cancer) and C6 (Rat Brain Tumor) cell lines at four different concentrations (5, 25, 50, 100 μg/ml) and compared with 5-Fluorouracil (5-FU), which is used as an anticancer drug.en_US
