dc.description.abstractÖZET Bu çalışmada, Sağlık Bakanlığı Urfa Harran Kapı Sağlık Ocağına başvuran ve deri leyişmaniyozisli olduğu düşünülen 21 kişinin, lezyonlanndan alman örnekler parazitolojik yönden incelendi. Lezyonlardan `lansede örnek alma` adım verdiğimiz yöntemle örnekler alınarak direkt mikroskobik incelemeleri yapıldı. Aynı örneklerin NNN ve sığır kanlı besiyerlerine ekimleri yapılarak kültürlerde promastigotlann üremelerine trimethoprim-sulfamethaxozole ve ofloksasinin etkileri araştırıldı. Direkt mikroskobik incelemede, toplam 21 örneğin 11 'inde (%52) amastigot görülürken, NNN besiyerlerine yapılan ekimlerin 15'inde (% 71) promastigotlann ürediği saptandı. Sığır kanlı besiyerinde ise üreme gözlenmedi. İn vitro koşullarda farklı derişimlerdeki trimethoprim-sulphametaxozole (Bactrim) (1, 2, 4, 8, 16 u,g/ml) ve ofloksasinin (Tarivid) (5, 10, 20, 40 ug/ml) NNN besiyerindeki promastigotlar üzerine etkili olmadıkları saptandı. 45
dc.description.abstractSUMMARY Comparison of direct and culture methods in the diagnosis of cutaneous leishmaniasis and the investigation of the effects of both trimethoprim- sulfamethaxozole and ofloxasin on promastigots In this study, samples taken from the lesions belonging to 21 people, who are thought to be cutaneous leishmaniosis and admitted to Şanlıurfa Harrankapı Health Center ruled by Ministry of Health, has been examined parasitologjcally. A direct microscobic examination has been done by the method called `taking sample by lancet`. By inoculating the same samples in NNN and bovine blood medium, the effects of trimethoprim-sulfamethaxozole and ofloxasin on growth promastigots in cultures have been investigated. While amastigots have been found in 1 1 (52%) of 21 samples in direct microscobic examination, growth of promastigots was observed in 15 (71%) cultures of the same samples. However, no growth has been observed in bovine blood medium. It is determined that trimemoprim-sulfamediaxozole (Bactrim) (1, 2, 4, 8, 16 ng/ml) and ofloxasin (Tarivid) (5, 10, 20, 40 ^ig/ml) in different concentrations have no effect on promastigots in NNN media under in vitro conditions. 46en_US
