dc.description.abstractBu çal mada, Türkiye'deki 27 do al alabal k populasyonunun mikroco rafik vemakroco rafik mikrosatelit DNA varyasyonu ara t r lm t r. Toplam be mikrosatelit lokusubu amaçla analiz edilmi tir. Çal lan alabal k populasyonlar nda lokuslar n tümü polimorfikolup ortalama alel say s 7.4 olarak bulunmu tur. Alabal k populasyonlar n n ço unda tek biralel saptanm ve ara t r lan be lokus için ortalama gözlenen heterozigotluk 0.254 olaraktespit edilmi tir. ,ncelenen populasyonlar n önemli bir k sm n n Hardy-Weinberg dengesindeolmad görülmü tür. Mikroco rafik alanda ayn havza ya da nehir sisteminde bulunanpopulasyonlar n da kendine özgü alelleri oldu u saptanm t r. Ayr ca, populasyonlar n iki erlikar la t r lmas sonucunda elde edilen FST de erlerinin ço u istatistiksel olarak önemlibulunmu tur. Makroco rafik alanda farkl havza sisteminde bulunan populasyonlar aras nda% 45.78 oran nda varyasyon tespit edilmi tir. Ayr ca daha önce mitokondri DNA'sanalizleriyle ortaya konulan Türkiye'deki alabal klar n n mitokondri soy hatlar , mikrosatelitDNA belirteçleri analizi ile de desteklenmi tir. Ancak mtDNA verilerine göre Tigris (TI) soyhatt na ait olan Çatak populasyonu, mikrosatelit DNA veri analizlerine göre Adriatic (AD)soy hatt içinde olan F rat Nehri'nin populasyonlar yla birlikte kümelenmi tir. MikrosatelitDNA ve mtDNA verileri, alabal klar n AD mitokondri soy hatt ile TI mitokondri soy hattaras nda muhtemel ikincil temaslar n oldu una i aret etmektedir.Anahtar Kelimeler: Salmo trutta, Mikrosatelit DNA, Anadolu, mitokondri DNA, alabal k vebiyoco rafya.
dc.description.abstractIn the present study, micro- and macro-geographic microsatellite DNA variations wereinvestigated in twenty seven brown trout populations in Turkey. A total of five microsatelliteloci were analyzed for this purpose. All the loci were polymorphic and average number ofalleles were 7.4 in the brown trout populations studied. Only a single allele was found in themost of the brown trout populations and average observed heterozygosity were 0.254 for thefive loci in question. Most of the populations did not show in the Hardy-Weinbergequilibrium. It was found that even populations in a micro-geographic region, same basin orriver system, had unique alleles. In addition, most of the Fst values obtained by the pairwisecomparison of the populations were statistically significant. Populations from different macro-geographic regions, different sea and river basins, 45.78% genetic variation was determined.Additionally, the mitochondrial DNA lineages of the brown trout in Turkey that havepreviously been identified by the mitochondrial DNA analyses were supported by the analysisof the microsatellite DNA markers. However, Çatak population which belongs to Tigris (TI)lineage was clustered together with the Euphrates populations within the Adriatic (AD)lineage according to the data analysis of the microsatellite DNA. Data of the bothmitochondrial DNA and microsatellite DNA indicated the possible secondary contactsbetween AD and TI lineages of the brown trout.Key Words: Salmo trutta, Microsatellite DNA, Anatolia, mitochondrial DNA, brown trout,biogeography.en_US
