ÖZET Nitrosolu bileşiklerden Dietilnitrosamin, N-Nitrosomorfolin ve N-Nitrosopirolidin 'in Na+/K+- ATPaz aktivitesini, zamana karşı in vivo olarak ne şekilde etkilediği çalışıldı. Bu amaçla, farelere bu bileşiklerin LD30 dozları olan 100 mg/kg, 168 mg/kg ve 360 mg/kg i`p. olarak enjekte edilip, 96. saate kadar enzim aktiviteleri izlendi. Dietil nitrosamin 'in enzimi kontrollere kıyasla % 77 ora nında aktive ettiği gözlendi. N-Nitrosomorfolin ile % 30, N-Nitrosopirolidin ile ise, % 22 oranın da inhibisyon elde edildi. Enzimi en fazla inhibe eden N-Nitrosomorfo lin 'in saf domuz serebral korteks Na+/K+ ATPaz'ı üzerindeki in vitro etkileri de incelendi. N-Nit rosomorfolin * in enzimi % 50 inhibe eden derişimi 27 mM olarak saptandı. Zamana göre inhibisyonun arttığı ve Sığır Serum Albümin 'inin inhibisyonu azalttığı gözlendi. înhibitörün enzime bağlandığı dializle gösterildi. İnhibisyon kinetikleri ince lenerek, N-Nitrosomorfolin* in Na /K+ ATPaz'ı52 nonkompetetif olarak inhibe ettiği bulundu. Mikrozomal sistemle metabolize edilen N-Nit- rosomorfolin 'in Na+/K+ ATPaz'ı inhibitörün kendi sinden fazla inhibe ettiği gösterilmekle birlikte metabolitlerle ilgili daha ayrıntılı bir çalışma nın gerekliliği vurgulandı.

SUMMARY The Effects of Some Nitrosamines on the Mouse Liver Na+/K+-ATPase Enzyme and their Binding Characteristics In this work, it has been attampted to in vestigate how N-Nitrosodiethylamine, N-Nitrosomor- pholine and N-Nitrosopyrrolidine, all nitroso com pounds, had an in vivo effect on the ATPase acti vity within a definite period of time. For this purpose, 100 mg/kg, 168 mg/kg and 360 mg/kg, LD_Q doses of these compounds, were injected to mice intraperitoneally and their enzymatic activities followed up until the 96 - hour. In comparison with the controls, it was observed that N-Nitroso diethylamine led to a 77 % activation of the en zyme. The inhibition brought about with N-Nitroso- morpholine was 30 % and that obtained with N-Ni trosopyrrolidine 22 %. The in vitro effects on the pure porcine cer ebral cortex Na+/K+-ATPase of N-Nitrosomorpholine inhibiting the enzyme most have also been investi-54 gated. The N-Nitrosomorpholine concentrations which had a 50 % inhibition of the enzyme was de termined to be 22 mM. It was observed that the in hibition increased during the course of time and Bovine Serum Albumin decreased the inhibition. By dialysis, it was demonstrated that the inhibitor was bound to the enzyme. By studying the inhibi tion kinetics, it was established that N-Nitroso morpholine inhibited Na /K -ATPase noncompetitive- iy. Although, it has been demonstrated that N-Ni trosomorpholine metabolized by the microsomal sys tem inhibited Na+/K+-ATPase more than the inhibi tor itself, the need for a more detailed investi gation on the metabolites has been indicated with emphasis.en_US
