dc.description.abstract34 ÖZET Bu araştırmada, farelere gebeliğin 14. gününden başlanarak 5 gün süre ile 6 ug günlük dozda (5x6ug) mısır yağmda çözündürülen DES oral yoldan uygulandı. îlaçla doğrudan etkileşen grup ve bunların dişi yavrularından kemik iliği alınarak kromozomlar incelendi ve(2n=40) adet ve akrosentrik yapıda oldukları görüldü.. Ayrıca Msp I enzimi ile kromozomlarda metillerime bölgeleri saptandı. Doğrudan DES uygulanan farelerin kromozomlarında hiperdiploidi,hipodiploidi ve toplam sayısal anomalilerde, ayrıca tek kromatidin telomerden birleşmesi, sentromerden birleşme ve toplam yapısal anomalilerde anlamlı artış izlendi. Bu farelerin dişi yavrularında ise daha az anomaliye rastlandı. Doğrudan ilaçla etkileşen grupta mikroçekirdek ve anafaz köprüsü bulgusu dikkat çekti. Her iki grupta da, mitotik indekste artış izlendi. Msp I- RE's uygulanan anne ve yavru denek farelerin kromozomlarında G bant benzeri yapılar oluştuğu saptandı. Doğrudan ilaçla etkilenen farelerde anomalilere sık rastlanması, DES etkisinin doz ve uygulama süresi ile bağlantılı olabileceği şeklinde yorumlanmıştır.
dc.description.abstract35 SUMMARY INVESTIGATION ON THE CHROMOSOMAL EFFECTS OF DES IN PREGNANT MICE AND SECOND GENERATION FEMALE OFFSPRINGS AND METHYLATION REGIONS, USING Msp I ENZYME In this research work, DES dissolved in corn oil was applied orally to mice in daily dosages of 6 pg (6X5 ug) for five days beginning on 14 th day of pregnancy. Bone marrow was obtained from the group directly affected by drug and the female offsprings, their chromosomes were examined and it was observed that these were n= 40 in number and acrocentric in structure. In addition, methylation regions of the chromosomes were determined by Msp I enzyme. Hyperdiploidy and hypodiploidy were observed in chromosomes of mice subjected to DES directly and significant increase in total numerical anomalies, single cnromatidine telomere association, centromere assosiation and total structural anomalies were noted. Fewer anomalies were encountered in the female offsprings of these mice. In the group directly affected by medicine, the increase in the number of the micronuclei and the anaphase - bridge finding attracted attention. These findings were not encountered in the female offsprings of mice. An increase of mitotic index in both groups was noted. Msp I RE's was applied on the chromosomes of mother mice and their offsprings used as experimental material. It has been observed that a G band- like structure had formed. In conclusion, anomalies frequently encountered in the mice directly subjected to drug suggested a relationship between DES effect and dosage and application period.en_US
