dc.description.abstractBu araştırmada Sivas ilinin farklı yerlerinden alınan 100 adet beyaz peynir örneğinde Enterohemorajik E.coli O157:H7 suşunun varlığı araştırıldı. Peynir örnekleri ön zenginleştirme işleminden sonra 4-metilumbelliferil-ß-D-glukuronid (MUG) içeren Lauryl Sulfat Triptoz Buyyona (LSTB) ekildi. Tümünde fekal E. coli ürediği saptandı. Sorbitol Mac Conkey agara yapılan ekimler sonucunda 22 örneğe ait plakta renksiz koloniler gözlendi. Bu kolonilerin mikroskobik incelemesi, indol, metil red, vogeus proskauer ve sitrat testleri yapıldı. İncelenen 22 örneğin 3'ünün test sonuçları E.coli O157:H7 ile uyumlu bulundu. Üç örneğe ait kolonilere lateks aglütinasyon testi yapıldı ve aglütinasyonun oluşmadığı görüldü.Sonuç olarak, toplam 100 adet peynir örneğinin tamamından fekal E.coli izole edilirken, E.coli O157:H7 suşu izole edilemedi. Satılan peynirlerin fekal E.coli gibi hijyen indikatörü mikroorganizmayla kontamine olması nedeniyle Türk Gıda Kodeksi Mikrobiyolojik Kriterler Tebliği'ne uymadığı ve peynirlerin tüketici sağlığı açısından risk oluşturabileceği kanısına varıldı.Anahtar kelimeler: Peynir, E.coli O157:H7
dc.description.abstractIn this study, the existence of enterohemorrhagik E.coli 0157:H7 strain in 100 white cheeses taken from different parts of Sivas was investigated. Cheese samples were inoculated to Lauryl Sulfate Triptoz Buyyon (LSTB) including 4-metilumbelliferil-ß-D-glukuronid (MUG) after the pre-enrichment procedure. It was observed in all that fecal E.coli was reproduced. Colourless colonies were observed in the plaques of 22 samples after the plantations applied to Sorbitol MacConkey Agar. These colonies? microscobic analysis, indol, methyl red, vogeus proskauer and sitrat tests were made. The three test results, which were examined, of 22 samples were found coherent with the E.coli. Latex agglutination test was made to the colonies belonging to these three samples and it was observed that agglutination was not formed.As a result, while fecal E.coli were isolating from all the 100 cheese samples, E.coli O157:H7 strain was not isolated. It was agreed that as the cheeses, which were sold, were contaminated with the hygen indicator microorganism such as fecal E.coli, they were not accord with the Turk Food Codex Microbiologic Criteria Notification and these cheeses will have risk for the health of consumers.Key words: Cheese, E.coli O157:H7en_US
