dc.description.abstractMissed abortus ve istemli gebelik evokuasyonu yapılan olguların desidua ve plasenta örneklerinde, birçok hücresel faaliyette rol oynayan TNF-? (Tümör Nekroz Faktörü-alfa), TGF-ß2 (Tümör Büyüme Faktörü-beta 2), IGF-I (İnsülin-benzeri Büyüme Faktörü-I), POMC (Pro-opiyomelanokortin), FOXO3A (Forkhead box O3A) ile proprotein konvertaz ailesi üyelerinden PC4 (Proprotein Konvertaz 4), PC6 (Proprotein Konvertaz 6), PC7 (Proprotein Konvertaz 7) ve FURİN moleküllerinin dağılımlarını immonohistokimyasal teknikle karşılaştırarak, missed abortus etiyolojisindeki etkileri araştırıldı. Çalışmaya Celal Bayar Üniversitesi Kadın Hastalıkları ve Doğum bölümüne başvuran, gebeliğin sonlandırılması kararı verilmiş missed abortus tanılı ve 10 hafta altı gebelik evokuasyonu istemi ile başvuran 10'ar hasta alındı. Her iki hasta grubundan endometriyal ve plasental doku örnekleri klasik doku takibi yöntemi ile bloklandı ve seri kesitleri alındı. Materyaller iki gruba ayrılarak incelendi. Seri kesitlerin birinci grubuna IHC uygulandı ve TNF-?, TGF-ß2, IGF-I, POMC, FOXO3A, PC4, PC6, PC7 ve FURİN primer antikorları ile immünohistokimyasal boyamalar uygulandı. Boyanan hücre sayısı H-Skor tekniği ile hesaplandı. Bu çalışmada, missed abortus olgularına ait desidua ve plasenta örneklerinde hücre proliferasyonu, invazyonu, migrasyonu, farklılaşması ve sağ kalımında rol oynayan TNF-?, TGF-ß, IGF-I, FOXO3A faktörlerinin artışı ve bu faktörlerin öncül formlarının posttranslasyonel modifikasyon yolu ile aktif formuna dönüşmesinde rol oynayan POMC, PC4, PC6 ve FURİN artışı ile paralellik göstermesi, bu faktörlerin fetal yaşam son bulduğu halde düşüğün gerçekleşmesini engellediğini düşündürdü.

The number of stained cells was calculated using H-Score technique. In this study, it is concluded that increment of TNF-?, TGF-ß, IGF-I, FOXO3A factors which play important roles in proliferation, invasion, migrasion, differentiation and survival of cells in the decidua and placenta samples, and paralleling increment of POMC, PC4, PC6 and FURİN which have roles to translate these factors into active forms from precursors via posttranslational modifications, reason of proceeding decidualization, placentation and preventing from abortion in spite of concluding the fetal life.en_US
