dc.description.abstract | Bu çalışmanın amacı, KBY'de üremik toksinlerin (üre, kreatinin, ürik asit, kalsiyum, fosfor) ve hemodiyaliz tedavi süresinin apoptozis üzerine etkisini, sitokeratinlerin apoptosiz sırasında kaspazlar tarafından yıkılması ile oluşan M30 biyobelirtecinin plazma konsantrasyonunu ölçerek araştırmaktır.Çalışmaya hemodiyaliz tedavisi alan yaşları ortalama 55 ± 14 olan 34 kadın ve 36 erkek toplam 70 hemodiyaliz hastası ve yaşları ortalama 45 ± 4 olan 11 kadın 9 erkek toplam 20 sağlıklı gönüllü dahil edildi. Hemodiyaliz hastaları tedavi sürelerine göre gruplandırıldı; 1.grup; kontrol grubu, 2.grup; 6 ay-3 yıl, 3.grup; 4-8 yıl, 4.grup; 8-10 yıl süreyle hemodiyaliz tedavisi görenler.Hemodiyaliz öncesi alınan kanlardan M30 düzeyi ELISA yöntemi ile üre, kreatinin, ürik asit, fosfor ve kalsiyum düzeyleri spektrofotometrik yöntem ile ölçüldü.Plazma M30 düzeyleri hemodiyaliz hasta gruplarında (2.grup; 189 ± 71,7 U/L, 3.grup; 182 ± 65,2 U/L, 4.grup; 288 ± 255,7 U/L) kontrol grubuna (122 ± 20,9 U/L) göre anlamlı oranda yüksek bulundu. (p<0,01) 2.grupta ürik asit ile M30 (p<0,05, r: 0,449), 3.grupta üre ile M30 (p<0,01, r: -0,521), 4.grupta ürik asit ile M30 (p<0,01, r: 0,590) arasında korelasyon vardı. Tüm hemodiyaliz hastalarında ise M30 ile ürik asit (p<0,05, r:0,291), fosfor (p<0,05, r: 0,265) ve kalsiyum (p<0,05, r: -0,253) arasında korelasyon bulundu.Sonuç olarak çalışmamızın bulguları KBY'li hastalarda apoptozisin arttığını ve bu artışta fosfor ve ürik asitin rolünün bulunabileceğini, hemodiyaliz tedavi sürelerinin apoptozis üzerine etkili olmadığını göstermiştir. | |
dc.description.abstract | The objective of this study is to investigate the impact of both the uremic toxins (urea, creatinine, uric acid, calcium, phosphorus) and the hemodialysis treatment duration onto apoptosis process by measuring the level of M30 biomarkers which are generated by the caspase?s cleave of cytokeratins during this apoptosis process.Seventy volunteer patients that were composed of 34 female and 36 male patients who had undergone hemodialysis treatment with a mean age 55+14 and twenty healthy volunteers that were composed of 11 female and 9 male with a mean age 45 + 4 were included to this study. Hemodialysis patients were grouped on the basis of their treatment duration; 1st group was taken as a control group, 2nd group was selected between the ranges of 6 months-3 years; 3rd group was between the ranges of 4-8 years; 4th group was between the ranges 8-10 years of hemodialysis treatment.Whilst M30 levels were measured via ELISA methods, urea, creatinine, uric acid, phosphorus and calcium were obtained via spectrophotometric methods in blood samples which were taken prior to hemodialysis treatment.As a result, M30 levels of plasma (2nd group: 189 ± 71,7 U/L, 3rd group; 182 ± 65,2 U/L, 4th group; 288 ± 255,7 U/L ) were found significantly higher in hemodialysis patient groups when compared to the control group. (p<0,01) The correlations among M30 and uric acid in 2nd group (p<0,05, r: 0,449); M30 and urea in 3rd group (p<0,01, r: -0,521); M30 and uric acid in 4th group (p<0,01, r: 0,590) were found statistically significant. Additionally, the correlations were found statistically significant among M30 levels and uric acid (p<0,05, r: 0,291), phosphorus (p<0,05, r:0,265), calcium (p<0.05, r: -0,253) in whole hemodialysis patients.In conclusion, we suggest that apoptosis increases in hemodialysis patients and is closely correlated with serum phosphorus and uric acid levels but total duration of hemodialysis is not correlated with the apoptosis. | en_US |
