dc.description.abstractÇalışmamızda 2.3 dihidro-7h-cycloheptan [1, 4]-dioxin'in (AD1 olarak kodlandırılan) antiinflammatuar aktivitesi sıçanlarda karragenin inflamasyon modelinde araştırıldı. Ayrıca, AD-1'in kapiller damar permeabiliteye etkileri hyaluronidaz testinde incelendi. AD-1 50,100 ve 200 mg/kg dozlarda karragenin inflamasyonunu sırası ile % 37 (p<0.05), % 71 (p<0.02) ve % 85 (p<0.001) olarak azaltmış, indometazin ise 25 mg/kg dozda karragenin inflamasyonunu % 67,9 (<0.02) oranında baskılamıştır. AD-1, 50 mg/kg dozda tavşanlarda hyaluronidazla artırılan kapiler damar permeabiliteyi 5. ve 30. dakikalarda kontrole göre anlamlı azaltmıştır. 25 mg/kg dozda kullanılan indometazin de hyaluronidazın kapiller permeabilite üzerindeki etkisini anlamlı olarak inhibe etmiştir. Çalışmamız, AD-1'in daha yüksek dozlarda (100 ve 200 mg/kg) karragenin inflamasyonunun baskılanması ve hyaluronidazla artırılan kapiller damar permeabilitenin azaltılmasında indometazinden daha etkili olduğunu göstermiştir. Ayrıca, AD-1 toksisitesinin indometazinden daha düşük olduğu görülmüştür.
dc.description.abstractIn our study, anti-inflammatory activity of 2.3 dihidro-7h-cycloheptan [1, 4]-dioxin (coded as AD1) was investigated in rats using carrageenan inflamation model. In addition, the effects of AD1 on capillary permeability were analyzed by hyaluronidase test. 50, 100, 200 mg/kg doses of AD-1 reduced carrageenan inflamation by 37 percent (p<0.05), 71 percent (p<0.02) and 85 percent (p<0.001) respectively. Where as 25 mg/kg dose of indomethacine inhibited carrageenan inflamation by 67.9 percent. AD-1 at 50 mg dose, significantly reduced hyaluronidase-induced capillary permeability at 5th and 30th minutes relative to controls in rabbits. Similarly, 25 mg/kg dose of indomethacine significantly inhibited the effect of hyaluronidase on capillary permeability. The present study demonstrated that, higher doses of AD-1 (100 and 200 mg/kg) were more effective in inhibiting carrageenan inflamation and in reducing hyaluronidase-induced capillary permeability than indomethacine. It was also observed that, toxicity of AD-1 was lower than that of indomethacine.en_US
