EŞİLHİSAR YÖRESİNDEKİ KOYUN VE KEÇİLERDEKİ BABESIAETKENLERİNİN REVERSE LINE BLOTTING (RLB) YÖNTEMİYLEARAŞTIRILMASIÖZETBu çalışma Yeşilhisar yöresinde Reverse Line Blotting (RLB) testi ve mikroskobik bakı ilekoyun ve keçilerdeki Babesia türlerinin prevalansını tespit etmek amacıyla yapılmıştır. Buamaçla Yeşilhisar merkez ve köylerinden rastgele seçilen sağlıklı 200 koyun ve 100 keçi olmaküzere toplam 300 küçük ruminanttan EDTA' lı tüplere kan alınmıştır. Kanlar RLB testi ile Catch-all, Babesia ovis ve Theileria ovis yönünden incelenmiştir. Aynı zamanda bu hayvanlardanperifer kan frotileri hazırlanarak mikroskobik muayenesi yapılmıştır.Mikroskobik muayene ile 200 koyunun 28'inde (% 14) ve 100 keçinin 5'inde (% 5) toplam 300hayvanın 33'ünde (% 11) Piroplasma pozitif saptandı. Bunlardan 9 (% 3) tanesinde Babesia spp.ve 24 (% 8) tanesinde de Theileria spp. tespit edilmiştir.RLB ile toplanan 200 koyun kan numunesinin 99'unda (% 49,5) ve 100 keçi numunesinin18'inde (% 18) toplamda 300 kan numunesinin 117 tanesinde (% 39) catch-all pozitifliksaptanmıştır. Catch-all pozitif kanların 4 tanesinde (% 1,33) Babesia ovis tespit edilirken, 106tanesinde (% 35,33) Theileria ovis ve 7 tanesinde (% 2,34) de T.ovis+B.ovis (miks enfeksiyon)tespit edilmiştir. B.motasi ve B.crassa'ya rastlanmamıştır.Anahtar Kelimeler: RLB, Prevalans, Catch-all, Piroplasma, Babesia ,
N OF THE BABESIA AGENTS IN SHEEP AND GOATS BY THEREVERSE LINE BLOT HYBRIDIZATION METHOD AROUND YEŞİLHİSARABSTRACTThis study was carried out on sheep and goats to detect Babesia spp. prevalence by Reverse LineBlotting (RLB) test and microscopic examination around Yeşilhisar. In this purpose, the bloodsamples were randomly collected into the tubes with EDTA from a total of 300 healthy smallruminants that consist of 200 sheep and 100 goats in Yeşilhisar and vicinity. Blood samples wereinvestigated against catch-all, Babesia ovis and Theileria ovis using RLB test. In addition, peripheral blood smears were prepared and examined under a microscope. In the microscopic examination, piroplasm were determined in 28 (14 %) of 200 sheep, 5 (5 %)of 100 goats and totally 33 (11 %) of 300 small ruminants. It was also determined that 9 (3 %) ofthem had Babesia spp. and 24 (8 %) of them had Theileria spp.In 300 small ruminants, catch-all positivity was found in 99 (49,5 %) of 200 sheep and 18 (18 %)of 100 goats totally 117 (39 %) of 300 using RLB. T.ovis was determined in 106 (39 %) ofpositive catch-all blood samples while both T.ovis and B.ovis were found mix infected in 7 (2,34%) of them, B. ovis was found infected only in 4 (1,33 %) of them. B.motasi and B.crassa werenot detected in the samples.Key Word: RLB, Prevalance, Catch-all, Piroplasm, Babesiaen_US
