Diabetes mellitus`lu hastalarda glikozillenmiş hemoglobin ve karbonik anhidraz miktarlarının tespiti

Abstract

In this study, glycosylated carbonic anhydrase values incarbonic anhydrase solution purified by affinity chromatographyand glycosylated hemoglobin values were determinedfor 10 healty control subjects (10 males, 22-30yr of age) and 10 patients with diabetis mellitus (8femilles and 2 males, 20-40 yr of age) .Determinations of glycosylated hemoglobin and glycosylatedcarbonic anhydrase were carried out by the colorimetricmethod developed by Parker and co-workers. Theamount of hemoglobin in the hemolized and the amount ofprotein in the purified enzyme solution were determinedby cyanmethemoglobin method and by coomassie-blue methodrespectively. While the glycosylated hemoglobin valuewas found 0,80 ± O,lll;'mol fructose/gr.Hb (mean ± SD).10 -4Glycosylated carbonic anhydrase value was 3,03.10 +_Ll0,53.10 . mg. glucose/mg. protein.in the control group,these values were 2,29 ± 0,90 pmol fructose/gr. Hb. and6,38.10 - ± 0,53.10 -4 mg. glucose/mg. protein (mean + SD)in diabetic group respectively.The results of statistical analyse showed that the differencebetween glycosylated hemoglobin and glycosylatedcarbonic anhydrase values for diabetic and control'groupswas important (for both PZO,OOl). I'lhile there was lessimportant correlation betvleen glycosylated hemoglobin andglycosylated carbonic anhydrase (r= -0,64, P~0,05) forthe control group, there was no correlation between thesevalues for the diabetic group (r= 0,49, P > 0, 05) •
