dc.description.abstractDihidropirimidin dehidrogenaz [5,6-dihidroursil: NADP+ oksidoredüktaz, EC 1.3.1.2; DPD] enzimi, sığır karaciğerinden saflaştırıldı. Saflaştırma; homojenatın hazırlanması, amonyum sülfat çöktürmesi, afinite kromatografisi yöntemleriyle yapıldı. Sığır karaciğeri DPD enziminin saflaştırılması %12,5 verimle 1660 kat olarak gerçekleştirildi.Ayrıca, sığır karaciğeri DPD enzim aktivitesi üzerine bazı ilaçların etkileri araştırıldı. İnhibisyon etkisi gösteren ilaçlardan oksitetrasiklin, siproflaksasin, kanamisin, ivermektin, enroflaksasin, seftazidim, sefoperazon, gentamisin, amikasin, ornidazol, metronidazol, sefuroksim, sefepim, ampisilin amoksisilin, meropenem, asiklovir, sefazolin ve penisilin G için I50 değerleri sırasıyla 0,030 mM, 0,046 mM, 0,080 mM, 0,120 mM, 0,130 mM, 0,140 mM, 0,610 mM,1,260 mM, 1,820 mM, 2,500 mM, 3,600 mM, 4,330 mM, 4,370 mM, 4,920 mM, 6,300 mM, 6,710 mM, 10,960 mM, 13,900 mM, ve 51,330 mM, Ki sabitleri de sırasıyla 0,050 mM, 0,090 mM, 0,109 mM, 0,117 mM, 0,123 mM, 0,130 mM, 0,185 mM, 1,020 mM, 2,010 mM, 2,096 mM, 2,115 mM, 2,700 mM, 3,730 mM, 5,240 mM, 9,570 mM, 9,605 mM, 9,710 mM, 17,200 mM ve 22,030 mM olarak hesaplandı.Anahtar Kelimeler: Dihidropirimidin dehidrogenaz, sığır, karaciğer, ilaç, inhibisyon
dc.description.abstractDihydropyrimidine dehydrogenase [5,6-dihydrouracil: NADP+ oxidoreductase, EC 1.3.1.2; DPD] enzyme was purified from bovin liver. The purification was performed by preparation of homogenate, ammonium sulphate precipitation, affinity chromatography. Purification of bovin liver DPD enzyme was obtained with a yield of 12.5 % as 1660 fold, respectively.In addition, effects of some drugs on bovin liver DPD enzyme activity were investigated. I50 values and Ki constants were calculated for oxytetracycline, ciprofloxacin, kanamycin, ivermectine, enrofloxacin, ceftazidime, cefoperazon, gentamycin, amicasin, ornidazole, metronidazole, cefuroxime, cefepime, ampicillin amoxicillin, meropenem, acyclovir, cefazolin and penicillin G from drugs showing inhibition effects. I50 values were calculate as 0.030 mM, 0.046 mM, 0.080 mM, 0.120 mM, 0.130 mM, 0.140 mM, 0.610 mM, 1.260 mM, 1.820 mM, 2.500 mM, 3.600 mM, 4.330 mM, 4.370 mM, 4.920 mM, 6.300 mM, 6.710 mM, 10.960 mM, 13.900 mM and 51.330 mM, respectively. Ki constants were found as 0.050 mM, 0.090 mM, 0.109 mM, 0.117 mM, 0.123 mM, 0.130 mM, 0.185 mM, 1.020 mM, 2.010 mM, 2.096 mM, 2.115 mM, 2.700 mM, 3.730 mM, 5.240 mM, 9.570 mM, 9.605 mM, 9.710 mM, 17.200 mM and 22.030 mM for bovin liver DPD, respectively.Keywords: Dihydroprimydin dehydrogenase, bovin, liver, drug, inhibitionen_US
