dc.description.abstractÖZET Amaç: Reflü nefropatisinde, serbest oksijen radikalleri ve nitrik oksitin etkilerinin araştırılması. Materyal ve metod: Çalışma yirmi prepubertal erkek tavşanda yapıldı. Grup 1 kontrol grubu, Grup 2 VUR+steril idrar grubu, Grup 3 VUR+enfekte idrar grubu olarak planlandı. VUR oluşturmak için sağ intravezikal üreterin ön yüzü üreterin mesaneye girdiği yere kadar kesildi. Grup 3'e ait tavşanların mesanelerine VUR oluşturduktan sonra ml'de 105 mikroorganizma bulunan E. coli süspansiyonundan iki mi verildi. VUR oluşturulmasından 3 hafta sonra Grup 2 ve 3'e voiding sistoüretrogram çekilerek tüm tavşanlarda sağ VUR geliştiği gösterildi. Deneysel VUR oluşturmadan önce ve üç hafta sonra grup 2 ile 3'e 99mTC-DMSA sintigrafîsi yapılarak böbrek uptake'leri hesaplandı.Vezikoüreteral reflü oluşturulduktan üç hafta sonra tüm tavşanlar, nitrit tayini için kan alındıktan sonra sakrifiye edilip bilateral nefrektomi yapıldı. Böbrek dokusunda malondialdehit ve nitrit ölçümü, serumda nitrit ölçümü ve histopatolojik inceleme yapıldı. Sonuçlar ortalama ± standart sapma olarak verildi. İstatistiksel çalışmada Varyans Analizi ve Scheffe testleri kullanıldı. Bulgular
dc.description.abstractSUMMARY Purpose: This experimental study was performed to determine the effects of free oxygen radicals and nitric oxide in reflux nephropathy. Material and methods: Twenty prepubertal male rabbits were divided into three groups. Group 1 was control group, Group 2 was VUR+sterile urine group and Group 3 was VUR+infected urine group. To create reflux the roof of the right intravesical ureter was incised at the point where the ureter penetrates the bladder wall. In Group 3, two ml of E. coli suspension (105 E coli in 1ml) was injected into the bladder in addition to VUR procedure. Right VUR was demonstrated in group 2 and 3 by micturition cystouretrography three weeks after the VUR. To determine the renal uptake in groups 2 and 3 99mTc-DMSA scintigraphy was performed, before and three weeks after the VUR procedure. To determine serum nitrite levels blood samples were taken from all groups three weeks after the experiment, and then bilateral nephrectomy wasdone, malondialdehyde and nitrite levels were measured in renal tissue. Histopathologic study was also performed. The results are expressed as mean ± SD. ANOVA and Scheffe's tests were used for statistical analysis. To explain the role of nitric oxide in reflux nephropathy needs more studies.en_US
