dc.description.abstractAsetilkolinesteraz enzimi (AChE; E.C.3.1.1.7) insan eritrositlerinden CNBr ile aktive edilen Sepharose 4B-L-Tirozin-Takrin afinite kromatografisi ile 658 kat, %23,5 verimle saflaştırıldı. Enzim saflığı SDS-PAGE ile kontrol edildi ve tek bant bulundu (70 kDa )AChE enzimi için optimum pH, optimum iyonik şiddet, optimum sıcaklık, stabil pH sırasıyla 7,6, 0,3 M, 50°C, 8,0 olarak tespit edildi. Ayrıca AChI ve DTNB substratları için KM ve Vmax değerleri sırasıyla 0,11 mM, 0,031 mM ve 0,082 EÜ/ml, 0,16 EÜ/ml olarak hesaplandı. AChE enzim aktivitesi üzerine vankomisin HCl, amikasin sülfat, pantoprazol, sefuroksim, diltizem HCl, tenoksikam antibiyotikleri, essitalopram, mirtazapin, fluoksetin, trazodon HCl, sodyum valproat, risperidon antidepresanları, BHA ve BHT sentetik antioksidanları, 2,6-diizopropilfenol(propofol), 2,6-dimetilfenol, 2,6-ditertbutilfenol, lidokain, bupivakain anestezikleri, etil alkol, aseton ve DMSO organik çözücüleri gibi farklı maddelerin inhibisyon etkileri incelendi. In vitro inhibisyon etkisi gösteren maddeler essitalopram okzalat, mirtazapin, risperidon, fluoksetin, pantoprazol, 2,6-diizopropilfenol, 2,6-dimetilfenol, 2,6-ditertbutilfenol, lidokain, bupivakain, BHA, BHT, etanol, aseton, dimetilsülfoksit için IC50 değerleri sırasıyla 1,06 mM, 0,019 mM, 0,428 mM, 1,62 mM, 1,97 mM, 5,94 µM, 6,96 µM, 36,3 µM, 260 µM, 5 µM, 32,85 µM, 36,3 µM, 2,24 M, 0,51 M, 0,51 M olarak hesaplandı. Aynı maddeler için sırasıyla ortalama Ki sabiti 1,30 mM, 0,012 mM, 0,23 mM, 1,48 mM, 1,26 mM, 3,93 µM, 2,06 µM, 18,9 µM, 14,3µM, 13,4 µM, 30,9 µM, 17,35 µM, 1,28 M, 0,19 M, 0,37 M olarak hesaplandı. dc.description.abstractAcetylcholinesterase enzyme (AChE: EC 3.1.1.7) was purified 658 fold with 23,5% yield from human ertyhroctes by Sepharose 4B-L-tyrosine-tacrine afinity column chromatograhphy that activated with CNBr. The purity enzyme was checked by SDS-PAGE and one band was observed (70 kDa). For AChE enzyme optimal pH, optimal temperature, stabil pH was determined as 7.6, 0.3 M, 50°C, 8.0 respectively. Also KM and Vmax values was calculated for AChI and DTNB substrates 0,11 mM, 0,031 mM ve 0,082 EU/mL, 0,16 EU/mL. On AChE enzyme activity were investigated the inhibitory effects of different substances such as vancomycn HCl, amicasin sulphate, pantoprazole, cefuroxime, diltizem hydrochlorid, tenoxicame antibiotics, escitalopram oczalate, mirtazapine, fluoxetine, trazodone hydrochlorid, sodium valproate, risperidone antidepressants, BHA and BHT unnative antioxidants, 2,6-dimethylphenol, 2,6-diisopropylphenol, 2,6-ditertbutylphenol, lidocain, bupivacain anesthezics, etylalcohol, aceton and dimethylsulfoxide organic solvents. IC50 values were calculated as 1,06 mM, 0,019 mM, 0,428 mM, 1,62 mM, 1,97 mM, 5,94 µM, 6,96 µM, 36,3 µM, 260 µM, 5 µM, 32,85 µM, 36,3 µM, 2,24 M, 0,51 M, 0,51 M for escitalopram oxalate, mirtazapine, risperidone, fluoxetine, pantoprazole, BHA, BHT, 2,6-diisopropylphenol, 2,6-dimethylphenol 2,6-ditertbutylphenol, lidocain, bupivacain, etylalcohol, aceton and dimethylsulfoxide substances which showed in vitro inhibitory effects respectively. Average Ki values were calculated as 1,30 mM, 0,012 mM, 0,23 mM, 1,48 mM, 1,26 mM, 3,93 µM, 2,06 µM, 18,9 µM, 14,3µM, 13,4µM, 30,9µM, 17,35µM, 1,28M, 0,19M, 0,37M for same substances respectively.
