dc.description.abstractIII ÖZET Spektrofluorimetri biyolojik moleküllerin kantitatif çalışmalarına oldukça duyarlı bir yöntemdir. Sunulan çalışmada bu yöntemle, poli-hidroksietilmetakrilat [poli(HEMA)] ve modifiye edilen poli(HEMA) membran ve mikropartiküllerde bovin serum albumin (BSA) ve fibrinojen adsorbsiyon profilleri ve kinetiği incelendi. Poli(HEMA) polimerine iyonik komonomerlerden dimetilaminoetllmetakrilat (DMAEMA) ve akrilik asit (AA) ilave edilerek yapı daha hidrofilik hale getirildi. Metilmetakrilat (MMA) komonomerl ile yapıya daha hidrofobik karakter verildi. Her iki protein yapısında bulunan Tryptophan (Trp) uçların bağıl İntrinslk fluoresans şiddetleri ölçülerek membran ve mikropartiküllere adsorblanan protein miktarı ve çözeltideki inaktivasyon hızlan (saat-1) tayin edildi. Polimerik yüzeylere protein adsorbsiyonunda zaman, sıcaklık ve pH etkileri incelendi. İlk 20 dakika içinde hızlı ve tersinmez adsorbsiyon, takiben daha yavaş ve tersinir adsorbsiyon izlendi. BSA ortalama olarak DMAEMA içeren membran ve mikropartiküllere sırasıyla 1.67 ug/cm2 ve 32.50 ug/g miktar tutulurken, aynı oranda AA içeren yüzeylerde bu değerler sırasıyla 0.85 ug/cm2 ve 9.10 ag/g değerlere düşmektedir. DMAEMA içeren üstteki yüzeylere ortalama adsorblanan fibrinojen 1.59 ug/cm2 ve 50.14 ug/g ; AA içeren yüzeylerde ise 0.69 ug/cm2 ve 24.30 ug/g olarak hesaplandı. Modifiye edilmemiş poli(HEMA) membran ve mikropartiküllere bir miktar fibrinojen adsorblanırken, BSA adsorbsiyonu izlenmedi. BSA ve fibrinojen adsorbsiyonu direk polimerik yüzeyden tekrarlanabilirsonuçlar vermediğinden alternatif bir yöntemle sonuçlar kontrol edildi. Bu yöntemle polimerik yüzeylerdeki protein adsorbsiyonu Eu(PDA)33` kompleksinin ekstrinsik fiuoresansından takip edildi. İntrinslk ve ekstrinsik fluoresans şiddetlerinden hesaplanan nicel sonuçlar istatistiksel olarak karşılaştırıldı. Polimerik membran ve mikropartiküllere BSA ve fibrinojen adsorbsiyonu FTIR spektrumlan ile de kanıtlandı.
dc.description.abstractiv SUMMARY Spectrofluorimetry is a very sensitive technique quantitative investigations of biological molecules. The technique was used in this study to follow the adsorption profiles and kinetics of two proteins; bovine serum albumin (BSA) and fibrinogen, onto membranes or microparticles prepared from poly-hydroxyethylmethacrylate [poly(HEMA)] and modified forms of poly(HEMA). This polymer was modified by the use of dimethylaminoethylmethacrylate (DMAEMA) and acrylic acid (AA) as comonomers to obtain more ionic hydrophilic structure. It was also modified by the use of methylmetacrylate (MMA) comonomer to obtain more hydrophobic character. By measuring the changes in fluorescence of intensty of tryptophan (Trp) in the structure of both proteins, the inactivation rate of the solution species and the adsorbed amount onto the membranes and microparticles were determined. The changes in protein uptake of the polymeric surfaces with time, temparature and pH of the solution were measured. It was found that a fast and irreversible adsorbption occurs for the initial 20 minutes which is followed by a slow and reversible adsorption. The average BSA adsorption for the membranes and microparticles that contain DMAEMA was found to the 1.67 ug/em2 and 32.50 ug/g respectively. The average BSA adsorption for the membranes and microparticles that contain the same percentage of AA, on the other hand, was found to be much less, 0.85 ug/cm2 and 9.10 ng/g respectively. The average fibrinogen adsorption for the membranes and microparticles that contain DMAEMA was found to the 1.59 ug/em2 and 50.14 ug/g respectively. The average fibrinogen adsorption for the membranes and microparticles that contain the same percentage of AA, on the other hand, was found to be much less, 0.69 ug/cm2 and 24.30 ug/g respectively. The unmodified poly(HEMA) surfaces of membranes and microparticles showed almost no BSA and fibrinogen adsorbtion. Although the quantitative evaluation of the results were not reproducible an experimental evidence for the BSA and fibrinogen adsorption was obtained by direct excitation of the polimeric surfaces. Using the extrinsic fluorescence of Eu(PDA)33` complex in solution an alternate method was proposed for the detection of the protein adsorption.The results of intrinslk and exrinsik fluorescence ıneasurements were compared statlstlcally. The adsorptlon of BSA and fibrinogen onto the pollmerîc membranes and mlcroparticles were also shovra by FTIR spectra.en_US
