dc.description.abstractIll ÖZET Bu çalışmada, tekstil endüstrisinde kullanılan ve nişastayı parçalayan Bacillus subtilis a-amilazı(a-l,4-D glucan glucanohydrola se, EC. 3. 2. 1.1, endoamylase) çalışıldı. İki farklı ortamda a- amilaz üretilerek aktivitelerine bakıldı. Bu ortamlardan daha erken maksimum aktiviteye ulaşan ve daha kararlı bir aktivite gösteren Modifiye Andersson ortamı sıvı ikili-faz sistemine uygulandı. Modifiye ettiğimiz sıvı ikili-faz sisteminde Dextran yerine DEAE- Dextran yapısında olan Cytodex 1 mikrotaşıyıcıları kullanıldı. PEG 600 (Polyethylen glycol 600) ve Cytodex 1 mikrotaşıyıcıları içeren sıvı ikili-faz sisteminde alfa-amilazın PEG fazında tutuklandığı saptandı. Böylece hücreden arı enzim üretimi gerçekleş tirildi.
dc.description.abstractXV SUMMARY In this study alpha-amylase of B. subtilis was studied. B. subtilis a-amylase (a-l,4-D glucan glucanohydyolase, E.C.3.2.1.1 endoamylase) that hydrolyze starch, is used in textile industry. Alpha-amylase were produced in two different media and enzyme activities were assayed. Alpha-amylase reached maximum activity in Modified Andersson's medium earlier than the other one. Also, enzyme activity was more stable in Modified Andersson's medium. Therefore this medium was applied to aqueous two-phase system. The aqueous two-phase system we modified, Cytodex 1 microcarriers (DEAE dextran in structure) were used instead of Dextran in bottom phase. It was established that alpha-amylase were immobilized in PEG phase of the aqueous two-phase system, composed of PEG 600 (Polyethylen glycol 600) and Cytodex 1 microcarriers. Thus cell-free enzyme production was verified.en_US
