Kırmızı Türk şaraplarında bulunan resveratrolün kromatografik yöntemle tayini

Abstract

Trans-resveratrol (3,5,4'-trihidroksi trans stilben) ve türevleri birçok bitki içinde özellikle üzüm ve yer fıstığında daha yoğun bulunabilen bir fenolik bileşiktir. Trans-resveratrol oldukça kuvvetli bir antioksidan etkiye sahiptir ve bu yüzden uzun süredir fitoterapik etkileri araştırılmaktadır. Özellikle koroner kalp hastalıklarında tedavi edici etkisi olduğu kesin olarak bilinmektedir. Tıp dünyasında Fransız Paradoksu olarak bilinen olay trans-resveratrolün tedavi edici etkisi ile açıklanmaktadır. Son zamanlarda trans-resveratrol'ün antitümör etkileri araştırılmaktadır. Trans-resveratrolün analizi ile ilgili çok sayıda çalışma son 20 yıldır kromatografi alanındaki dergilerde yayınlanmıştır. Bu makalelerin çoğu üzümden elde edilen ürünlerde özellikle şaraplarda bulunan trans-resveratrol tayini üzerinedir ve genellikle HPLC ile tampon taşıyıcı faz ve gradient çalışmalardır. Yakın zamandaki makalelerde kütle dedektörü (MS) kullanılmıştır. Ekstraksiyon esaslı analiz çok az sayıda rapor edilmiştir. Bu çalışmada Türkiye'de üretilen bazı şarap örneklerinde trans-resveratrolün daha basit kromatografik analizi amaç seçilmiş ve basit ekstraksiyon teknikleri ile isokratik HPLC tekniği birleştirilmiştir. Bu amaçla 8 adet kırmızı Türk şarabı ekstraksiyon esaslı yöntem ile %60 MeCN: %40 EtOH taşıyıcı fazında analiz edilmiştir. Standart ekleme yöntemi ile analizin doğruluğu yorumlanmıştır. HPLC ile tekrarlanabilir nicel sonuçlar alınmıştır.

Trans-resveratrol (3,5,4?-trihydroxy trans stilbene) and its derivatives are phenolic compounds present in many plant species, especially in grape and peanut. Trans-resveratrol is a considerably strong antioxidant indeed. For this reason its phytotherapic effects are studied for a long time. It is known for sure that trans-resveratrol has a curing potency for coroner hearth decease. The incident known as French Paradox in medicine is explained with the curing effect of trans-resveratrol. The antitumor effects of trans-resveratrol have been researched lately. A number of studies about trans-resveratrol analysis have been published in the last 20 years The majority of these articles are about the trans-resveratrol analysis in products of grape, especially in wine. They are mostly based on gradient, buffer mobile phase experiments conducted with HPLC systems. In the recent studies, mass detector (MS) systems were employed. Only a few extraction based analysis methods have been reported. The purpose of this study is to develop a relatively simple chromatohraphic analysis method for trans-resveratrol in domestic wine samples from Turkey. Basic extraction techniques are combined with isocratic HPLC applications in order to do this. For this purpose, 8 Turkish red wines were analyzed in a mobile phase of %60 MeCN: %40 EtOH by a extraction based method. The accuracy of this method were investigated by standard addition method. Repeatable quantitative results were obtained with HPLC.
