Dr. Mehmet BABAOGLU KONYA-2003, Sayfa: 70 Jüri : Doç. Dr. Mehmet BABAOGLU : Prof. Dr. Sait GEZGİN : Prof. Dr. Bayram SADE Buğday (Triticum durum ev. Kızıltan-91) ve arpa (Hordeum vulgar e ev. Tokak-157/37) in vitro fidelerinde bor alımı ve çeşitli organlarda bor birkiminin dağılımı ICP-AES (Inductivelly Coupled Plasma-Atomic Emission Spectrometry- Varian Vista Model Axiel) kullanılarak araştırılmıştır. Ayrıca farklı bor konsantrasyonlanmn tohumların çimlenme yüzdeleri üzerine etkisi değerlendirilmiştir. Tohumlar; 200 ml'lik cam kavanozlarda, %0.7 agar, %3 sakkaroz ve sırasıyla; 0.0, 6.2, 18.6, 55.8, 111.6 mg/1 H3B03 (0.00, 1.05, 3.25, 9.76, 19.5 mg B/l) içeren 50 mi MS besin ortamında, her kavanozda 5 adet tohum olacak şekilde kültüre alınmıştır. Bor alımı ve birikiminin tespiti için, 20 günlük fidelerin kurutulmuş kök ve gövde kısımları (0.1-1 g) ve fidelerin yetiştiği besin ortamları (5.0-6.0 g) mikrodalgada (CEM-Mars x 5) 10 mi HN03 ile 170 PSI basınçta 200°C'de 40 dak. yakılmış ve numunelerde ICP-AES ile bor analizi yapılmıştır. Kızıltan-91 'in köklerindeki en düşük bor birikimi (2.2 ug B/l) bor konsantrasyonun 0 mg/1 H3BO3 olduğu ortamda kültüre alınanlarda bulunurken, en yüksek bor birikimi (15.1 ug B/l) en yüksek ortam bor konsantrasyonunda (111.6 mg/1 H3BO3) tespit edilmiştir. Benzer şekilde Kızıltan-91'in gövdesi tarafından en yüksek seviyede bor alımı (67.6 ug B/l) bor konsantrasyonunun en yüksek olduğu ortamda (111.6 mg/1 H3BO3) olmuştur. Kızıltan-91) and barley {Hordeum vulgare cv. Tokak-157/37) and distribution of B accumulation in various organs were investigated using and Inductively Coupled Plasma Spectrometer (ICP-AES, Varian Vista Model Axiel). Besides, the effect of varying B concentrations on germination rates of the seeds were evaluated. In vitro seedlings of wheat and barley were grown containing 50 ml of MS- agar medium (%0.7, w/v) with %3 (w/v) sucrose and 0.00, 1.05, 3.25, 9.76 and 19.5 mg/L boron corresponding to 0.0, 6.2, 18.6, 111,6 mg/1 boric acid (H3BO3) respectivelly. Seed germination rates as per boron concentration were recorded. Seedlings of which roots and shoots was removed used as samples for the analysis of boron uptake. Dried plant samples (0.1-1 g) and plant culture media (5.0-6.0 g) were transferred into plastic bottles containing 10 ml of 65 % (v/v) nitric acid (HNO3) solution. Samples were ashed at 200°C for 40 min in microwave (CEM-Mars x 5). A ICP-AES (Varian Vista Model Axiel) device was used for analyses followed by sample preperation in microwave. Increasing B concentrations did not provoke any clear effect on plant development of any species tested. The lowest concentration of B (2.2 ug B/l) accumulated was found in roots of Kızıltan-91 seedling cultured in medium containing 0 mg/1 H3BO3 whereas the highest B accumulation (15.1 ug B/l) was detected again in roots of Kızıltan-91 in medium containing the highest concentration of B (111.6 mg/1 H3BO3). Similarly, the highest level of B absorption (67.6 ug B/l) by the shoots of Kızıltan-91 took place in media containing the highest level of Boric acid (1 1 1.6 mg/1 H3BO3) with lowest B concentration was found in shoots at lowest medium B concentration, showing that there was a linear relation with respect of B content of the culture medium and B accumulation in the organs of Kızıltan-91.
