dc.description.abstractÖZET Bu çalışmada, sıçanlarda nasal mast hücrelerinin morfolojisi ve morfin sülfat ve kromolin sodyumun etkisi çeşitli boya metodları kullanılarak ışık mikroskobik düzeyde araştırıldı. Kullanılan 30 adet albino dişi ve erkek sıçan Selçuk Üniversitesi Tıp Fakültesi Deney Hayvanları Laboratuvarından temin edildi. Sıçanlar üç gruba ayrıldı. Kontrol grubundaki sıçanlar hiçbir madde verilmeden eterle bayıltıldı ve regio respiratoria nasi'den frontal kesiler alındı. İkinci gruba perkutan yoldan 200 mg/kg morfin sülfat, üçüncü gruba ise inhalasyon yolu ile kromolin sodyum verildi ve burunları kesildi. Kesilen burunların hepsi oda ısısında üç gün Carnoy fıksatifi için de tesbit edildi. Alkol takibi yapılan dokulardan 5 mikrometre kalınlığında kesitler alınarak toluidin mavisi ve alcian mavisi-safranin 0 ile boyanarak incelendi. Sonuçlar nasal mast hücrelerinin mukozal ve bağ dokusu mast hücreleri olmak üzere iki alt grubunun olduğunu, alcian mavisi-safranin 0 boyasının mukozal mast hücrelerim mavi, bağ dokusu mast hücrelerini ise kırmızı ve kırmızımtrak-mavi renkte boyadığım; toluidin mavisinin ise bütün mast hücrelerini koyu menekşe-mor boyadığım gösterdi. Morfin sülfatın stimule edici, kromolin sodyumun ise inhibe edici maddeler olduğu tesbit edildi.

SUMMARY LIGHT MICROSCOPIC INVESTIGATION OF STIMULATED AND INHIBITED NASAL MAST CELLS OF RAT USING HISTOCHEMİCAL METHODS In this light microscopic study, morphology of nasal mast cells and the effects of morphine sulfate and cromolyn sodium was investigated in rats using alcian blue-safranin O and toluidin blue staining methods. The investigation was performed on 30 male and female albino rats which were obtained from the Experimental Animal Laboratory of the Faculty of Medicine, Selçuk University. Rats were divided into three groups of ten rats each. First group was considered as control group. Percutaneous injection of 200 mg/kg morphine sülfat was administered to the second group and cromolyn sodi um was given to the third group by inhalation. Rats were anesthetized by ether and regio respiratoria nasi of rats were frontally excised. Tissues were fixed in Carnoy solution for three days at room temperature, dehy drated with absolute alcohol and embedded in paraffin. Five micrometre sections were cut from the paraffin blocks using rotary microtome and stained with alcian blue - safranin O and toluidin blue solutions. In conclusion, this study revealed that there are two distinct mast cell population in the rat nose: mucosal mast cells and connective tissue mast cells. Mucosal mast cells stained blue with alcian blue - safranin O solution while connective tissue mast cells stained red and reddish - blue. Both mast cell populations stained dark violet by toluidin blue solution. This study indicates that, as an inhibitory agent, cromolyn sodium is inef fective on nasal mucosal mast cells but significantly effective on connective tissue mast cells, on the other hand, morphine sulfate, as histamine - re leasing agent, is found to be effective on both mast cell population.en_US
