dc.description.abstractÖZET Bu çalışmada septisemili tavuklardan izole edilen 61 E. coli susu ile ishalli buzağılardan izole edilmiş 24 E. coli susu ile çalışıldı ve bu suşların seroüpik özellikleri ile pilus (K99, K88. F41, 987P, Tip 1) ve patojenite (enteropatojenite, enterotoksijenite, verotoksijenite) özellikleri incelendi. Tavşan ince barsak lup testinde buzağı orijinli suşlann % 33.3'ünün enteropatojenik, infantil fare testinde ise % 20.8'inin enterotoksijenik olduğu tesbit edildi. Vero hücre kültüründe yapılan verotoksijenite testinde ise suşlann % 29.1 'inin verotoksijenik karakterde olduğu gözlenmiştir. Tavuk orijinli suşlarda, ise, % 31.1 oranında enteropatogenite % 22.9 oranında enterotoksijenite ve % 34.4oranında verotoksijenite ile karşılaşıldı. Pilus tiplendirme çalışmaları sonucunda buzağı orijinli suşlann 8'inin K99, 4'ünün F41, l'inin K88 pilusu, 5 susun hem K99 ve hemde F41 pilusu taşıdığı, 1 susta ise K99, K88, F41 pilusunun birlikte bulunduğu tesbit edildi. Tavuk orijinli suşlarda ise 23 suşda K99, 29 susta F41, 2 susta da K88 fimbria varlığı belirlendi. Ancak 6 susta F41 ve K99 fîmbrianın birlikte bulunmaktaydı. İncelenen tavuk orijinli suşlann 01, 02, 078 antiserumlan ile % 63.9'unun somatik O grubu belirlendi. Genel olarak suşlann % 36. l'inin Tip 1 A fimbria'yı, % 19.7'sinin Tip 1 like fimbira'yı sentezlediği be lirlendi. Somatik antijenlerine göre tavuk orijinli suşlann 12'si 01,18 adedi 02, 1 adedi 078, 1 adedi 01/02, 5 adedi de 02/078 olarak se- rotiplendirilmiştir. 39
dc.description.abstractSUMMARY Studies on adhesirs (K99, K88, F41, 987P and type - 1 fimbriae) and enterotoxins (STa and VT) of E. coli strains isolated from calves with diarrhoea and hens with septisemiae. In this study, 61 E. coli strains isolated from hens with septisemia and 24 E. coli strains isolated from diarrhaeic calves were studied. Some pathogenic characters of these isolates such as pilus antijens (K99, K88, 987P, F41, type 1), enteropathogenic, enterotoxigenic and verotoxigenic properties were examined. It was determinated that 33.3% of E. coli strains from calves was enterotoxigenic by rabbit ileal loop assay and 20.8% was enterotoxigenic by infant mouse assay. By verotoxigenity test, 29. 1 % of isolates was found to be verotoxigenic. 31.1% of the 61 E. coli isolates from septisemic hens was enteropathogenic, 22.9% was enterotoxigenic and 34.4% was ve rotoxigenic. In term of pilus typing 8 strains of E. coli originated calves had K99, 4 strains had F41, 1 strain had K88 antigen, 1 strain had K99 + F41 + K88 and 5 strains had both F41 and K99 `antigens. The serotypes of 63.9 % E. coli strain originated from hens were determined using 01, 02 and 078 E. coli antisera. It were serotyped that 12 strains were 01 serotype, 18 strains were 02 serotype, 3 strains were 078 serotypes; 1 strains were 01/02 and 5 strains were 02/078 serotypes. It were serotyped that 36. 1% of the E. coli isolated originated from hens had typ 1A pilus/fimbria. But type 1 like pili were found 13.1 % of the strains 40en_US
