dc.description.abstractÖZET Keton cisimlerinden aseloasetal (Ar.Ac) ve 3-Hidroksibıitirat (3- HB) kanda yeni geliştirilen bir enzimatik metodla (ayin edildi. 3-HB tayin prensibi EDTA'lı total kan perklorik asitle deproteinize edildikten sonra 3-HB Dehidrogenaz (3-HBDH) varlığında 3-HB 'in NAD+ ile asetoasetata yükseltgenmesi ve oluşan NADH'ın diaforaz enzimi ile iyodonitrotetrazolyum klorürü(INT) renkli formazan bileşiğine dönüştürmesi esasına dayanır. Asetoasetat (AcAc) tayin prensibi ise asetoasetatın NADH ile birlikte 3-HBDH vasıtasıyla 3-HB ve okside NAD+'ye dönüşmesi reaksiyonuna dayanır. 3-HB düzeyleri 0.0123+0.0102 mmol/L (Aritmetik ortalama ± standart sapma) (medyan: 0.010mmol/L), AcAc düzeyleri 0.0344± 0.0228 (medyan: 0.0260 mmol/L) olarak bulundu. 3- HB tayininin hassasiyeti (within-run) düşük, orta ve yüksek konsantrasyonlu örneklerde %CV cinsinden 2 ile 10 arasında, AcAc için ise 1 ile 6 arasında bulundu. Dolaysıyla her iki metodun yeterli hassasiyette olduğu kanaatine varıldı. Bu tayinlerden asetoasetat tayini daha kısa süreli ve maliyeti düşük olduğu için diyabetik ketoasidoz takibinde kullanabileceği sonucuna varıldı.
dc.description.abstractSUMMARY Acetoacetate (AcAc) and 3-Hydroxybutyrate (3-HB), ketone bodies, were determined by a new enzymatic method in blood. Determination of 3-HB levels is based on the following principle: After a sample is deproteinized by perchloric acid, in the presence of the enzyme 3-hydroxybutyrate dehydrogenase (3-HBDH), D-3- hydroxybutyric acid (D-3-hydroxybutyrate) is oxidized by nicotinamide adenine dinucleotide (NAD+) to acetoacetic acid (acetoacetate). In the presence of diaphorase, the NADH formed converts iodonitrotetrazolium chloride(INT) to a formazon. The acetoacetate determination method is based on the following principle: Acetoacetate is converted to 3-HB by reduced nicotinamide adenine dinücleotide(NADH) in the presence of 3-hydroxybutyrate dehydrogenase (3-HBDH). The mean 3-HB level was found to be 0.0123+0.0102 mmol/L (the arithmetic mean ±standard deviation) (median : 0.010 mmol/L) and the mean AcAc level was 0.0344±0.0228 (median: 0.0260 mmol/L). While the precision of 3-HB assay was found to be 2 to 10 in terms of CV% for the samples of low, medium and high concentrations, AcAc valve was to be 1 to 6. Thus, it was concluded that each of the methods have enough precisions. The acetoacetate assay takes lower time and has lower costs. Therefore, acetoacetate may be used alone for following up diabetic ketoacidosis.en_US
