ÖZET Bu çalışmada, yetişkin peıiodontitisli 24 hastadan alman 51 ve 10 sağlıklı bireyden alman 20 subgingival plak örneği, Actinobacillus actinomycetemcomitans, Prevotella intermedia, Porphyromonas gingivalis, Bacteroides forsythus, Eikenella corrodens, Fusobacterium nucleatıtm, Peptostreptococcus micros, Streptococcus intermedins, Treponema denticola ve Campylobacter rectus'u içeren 10 periodontopatojen bakteri varlığı açısından PCR yöntemi ile incelendi. Direkt PCR incelemesi yapılan subgingival örneklerde C. rectus ve F. nucleatum %90, A. actinomycetemcomitans ve S. intermedius %62, P. gingivalis ve E. corrodens %43, B. forsythus %3Î, P. intermedia %29, P. micros %18 ve T. denticola °/<A oranında tespit edildi. Sağlıklı kontrol grubuna ait 20 subgingival örneğin %25'inde A. actinomycetemcomitans, %40'ında C. rectus ve F. nucleatum tespit edildi. PCR yönteminin özellikle kültürü zor ve zaman alıcı anaerobik periodontopatojen bakterilerin tanısında uygun bir yöntem olarak kullanılabileceği gösterildi.

SUMMARY RAPID DETECTION OF TEN SUSPECTED PERIODONTAL PATHOGENS IN SUBGINGIVAL PLAQUE SAMPLES FROM ADULT PERIODONTITIS PATD2NTSBYPCR In this study, 51 subgingival plaque samples from 24 subjects with adult periodontitis and 20 subgingival plaque samples from 10 healthy subjects were investigated by PCR for 10 periodontopathogen bacteria including Actinobacillus actinomycetemcornitans, Prevotella intermedia, Porphyromonas gingivalis, Bacteroides forsythus, Eikenella corrodens, Fusobacterium nucleatum, Peptostreptococcus micros, Streptococcus intermedins, Treponema denticola and Campylobacter rectus. In the adult periodontitis (AP), the most frequently detected organisms were C. rectus and F. nucleatum (%90), A. actinomycetemcornitans and S. intermedius (%62), P. gingivalis and E. corrodens (%43), B. forsythus (%31), P. intermedia (%29), P. micros (%18), T. denticola (%4), respectively. From 20 subgingival plaque samples from healthy subjects, only A. actinomycetemcornitans (25 %), F. nucleatum and C. rectus (40 %) were detected. PCR-based system for detecting subgingival microbial profile may help to diagnose and manage treatment of adult periodontitis cases and associated pathogens which are mostly anaerobes and difficult to culture.en_US
