dc.description.abstractAmaç: Geçici beyin iskemisi oluşturulan ratlardaki iskemi ve reperfüzyon hasarı üzerine seftriakson'un hipokampal bölgede malondialdehid düzeylerine ve nörolojik muayene üzerine olan etkilerinin araştırılması amaçlanmıştır. Materyal ve Metod: Çalısmamızda 220–280 g ağırlığında olan 38 adet spraque dawley sıçanı deneye alındı. İki damar oklüzyon + hipotansiyon iskemi-reperfüzyon modeli uygulaması için, bilateral karotis arterler geçici olarak (30 dk) kliplenip intrakardiak 3 cc kadar kan alındıktan sonra hipotansiyon yaratılmıştır. 30 dk sonunda klipler çıkarılıp reperfüzyon ortamı oluşturulmuştur. 12 sıçandan oluşan bir gruba 30 dk bilateral karotis kliplemesi + hipotansiyon (10ml/kg) yapıldıktan sonra her gün seftriakson 200 mg/kg/gün intraperitoneal verildi. 1., 4., 7. ve 10. günlerde nörolojik muayeneleri yapıldı. 10.günün sonunda sakrifiye edildi ve beyin dokusu çıkarıldı. 12 sıçandan oluşan diğer guruba 30 dk bilateral karotis kliplemesi+hipotansiyon (10ml/kg) oluşturularak sadece iskemi yapıldı (2016/54 nolu çalışma ile ortak grup). 1., 4., 7. ve 10. günlerde nörolojik muayeneleri yapıldı. 10. günün sonunda sakrifiye edildi ve beyin dokusu çıkarıldı. 7 rattan oluşan bir guruba sadece cilt laserasyonu yapıldı ve 200 mg/kg/gün intraperitoneal seftriakson verildi. 1., 4., 7. ve 10. günlerde nörolojik muayeneleri yapıldı. 10.günün sonunda sakrifiye edildi ve beyin dokusu çıkarıldı. 7 rattan oluşan son gruba sadece 15 dakika anestezi verildi. 1., 4., 7. ve 10. günlerde nörolojik muayeneleri yapıldı. 10.günün sonunda sakrifiye edildi ve beyin dokusu çıkarıldı.Bulgular: Beyin dokusundaki MDA seviyesi kontrol grubu ve iskemi grubu ile kıyaslandığında doku MDA seviyesini önemli miktarda arttırdığı belirlenmiştir (p<0.05). Pür kontrol ve seftriakson gruplarının MDA seviyeleri arasında anlamlı bir fark bulunamazken (p>0.05), iskemi grubu ile kıyaslandığında seftriakson uygulamasının anlamlı şekilde doku MDA seviyesini düşürdüğü tespit edilmiştir (p<0.05).Serum MDA seviyesi iskemi grubunun kontrol grubu ile kıyaslandığında önemli derecede arttığı gözlenmiştir (p<0.05). İskemide uygulanan seftriaksonun serum MDA seviyesinde belirgin bir düşüş sağlayamadığı belirlenmiştir (p>0.05).Sonuç: Serebral iskemi ve reperfüzyon hasarlanmasında seftriaksonun serumda artan MDA'yı düşürmediği fakat serebral dokuda MDA'yı düşürerek nöroprotektif etkinliğinin olduğu çalışmamızda gösterildi.Anahtar Kelimeler: iskemi, malondialdehid, seftriakson
dc.description.abstractPurpose: The aim of this study was to investigate the effects of ceftriaxone on malondialdehid in hypocampal region and the neurologic findings at rats with artificially maintained transient brain ischemia.Materials and Method: In our study, 38 spraque dawley rats weighing 220-280 g were tested. Two-vessel occlusion + hypotension for ischemia-reperfusion model application, bilateral carotid arteries were temporarily clipped (30 min) and 3 cc of blood was taken in to the intracardiac volume and hypotension was created.After 30 min, the clips were removed and a reperfusion medium was created. A group of 12 rats was done 30 min bilateral carotid artery clipping + hypotension (10 ml / kg) and after there was given intraperitoneal ceftriaxone 200 mg / kg / day dose every day. On the 1st, 4th, 7th and 10th days, neurological examinations were performed. At the end of the ten day, the rats was sacrificed and the brain tissue was removed. Other group consisting of 12 rats was done only 30 min bilateral carotid artery clipping + hypotension (10 ml / kg) and only was done ischemia (joint experiment group with the study of 2016/54). On the 1st, 4th, 7th and 10th days, neurological examinations were performed. At the end of the ten day, the rats was sacrificed and the brain tissue was removed. Other group consisting of 7 rats was done only skin laceration and received intraperitoneal ceftriaxone at 200 mg / kg / day. On the 1st, 4th, 7th and 10th days, neurological examinations were performed. At the end of the ten day, the rats was sacrificed and the brain tissue was removed. The last group consisting of 7 rats was given only 15 minutes anesthesia. On the 1st, 4th, 7th and 10th days, neurological examinations were performed. At the end of the ten day, the rats was sacrificed and the brain tissue was removed.Findings: It was determined that ischemia administration significantly increased brain MDA levels compared with brain tissue control group MDA level (p <0.05). While there was no significant difference between MDA levels of sham and ceftriakson groups (p> 0.05), it was determined that ceftriakson application decreased significantly MDA level compared to ischemia group (p <0.05). The MDA level of the ischemia group was significantly increased compared to the control group serum MDA level (p <0.05). While the difference between the control group and the sham group was not significant (p> 0.05), it was found that there was no significant decrease in ceftriakson serum MDA level in ischemia (p> 0.05).Results: In this study, statistically, it was determined that ceftriaxone has does not reduce level of MDA in the plasma, but the other hand statistically, it was determined that ceftriaxone has does reduce level of MDA in the brain tissue and it was showed neuroprotective effect.Key Words: ceftriaxone, ischemia, malondialdehiden_US
