dc.description.abstractIV ÖZET Salman, M.C., Pehik organ prolapsusunım doku düzeyinde değerlendirilmesi, Hacettepe Üniversitesi Tıp Fakültesi, Kadın Hastalıkları ve Doğum Tezi, Ankara 2004. Pelvik organ prolapsusu nedeniyle vajinal histerektomi yapılan hastalar ve myom gibi benign jinekolojik hastalıklar nedeniyle abdominal histerektomi yapılan hastaların kardinal ligamentlerinden alınan örnekler ışık mikroskobu ve elektron mikroskobunda incelendi. Hematoksilen-eosin, trikrom ve van Gieson boyamaları yapılarak ışık mikroskopi düzeyinde bağ dokusundaki fibroblastlann ve liflerin miktarına bakıldı. Elektron mikroskopi düzeyinde bağ dokusundaki kollajen liflerin ve fibroblastlann miktarına bakıldı, kollajen liflerin çaplan ölçüldü. Işık mikroskobik incelemede pelvik organ prolapsusu olan 10 hastanın 8'inde bağ dokusu komponentlerinde azalma saptamrken kontrol grubundaki 10 hastanın 7'sinde bağ dokusu normal bulundu. Elektron mikroskobik incelemede ise pelvik organ prolapsusu olanlarda kontrol grubu ile karşılaştınldığında fibroblastlarda azalma, kollajen konsantrasyonunda ve kollajen lif çapmda artış saptandı, menopozal durumun da burada etkili olduğu bulundu. Buna göre pelvik organ prolapsusu bağ dokusu değişiklikleri ile ilişkili bir hastalıktır ve mevcut bağ dokusu değişikliklerinin değerlendirilmesi en iyi şekilde elektron mikroskopiyle mümkündür. dc.description.abstractABSTRACT Salman, M.C., Tissue level evaluation of pelvic organ prolapse, Hacettepe University Faculty of Medicine, Thesis in Obstetrics and Gynecology, Ankara 2004. Cardinal ligament samples obtained from patients who underwent vaginal hysterectomy because of pelvic organ prolapse and abdominal hysterectomy because of benign disorders like myomas were examined by light and electron microscopy. After hematoxylin-eosine, trichrome and van Gieson staining the amount of fibroblasts and connective tissue fibers was evaluated in light microscopic level. In electron microscopic level, the amounts of fibroblasts and collagen fibers in connective tissue were examined and the diameters of collagen fibers were measured. In 8 of 10 patients with pelvic organ prolapse a decrease in connective tissue components was detected while the connective tissue was found to be normal in 7 of 10 patients in control group by light microscopic evaluation. The electron microscopic evaluation revealed that the patients with pelvic organ prolapse had decreased number of fibroblasts, increased collagen fiber concentration and diameter compared with control group, menopausal status was also found to be effective in these changes. Therefore, pelvic organ prolapse is a disease mat is related with connective tissue changes and these connective tissue changes can be best evaluated by electron microscopy.
