dc.description.abstractÖZETMeme kanseri 2006 T.C. saglık bakanlıgı verilerine görekadınlarda en sık görülen kanserdir. Amerika Birlesik Devletleri 2011verilerine göre her 8 kadından biri yasam boyu meme kanseri riski ilekarsı karsıya kalacaktır. MTHFR (Metilentetrahidrofolat redüktaz),5.10-metilentetrahidrofolat'ı , 5-metiltetrahidrofolat'a indirgeyen,homosistein'in metionin'e çevrilmesinde bir karbon donör olarak islevgören bir enzimdir. MTHFR ,DNA sentezi ,DNA tamiri vemetilasyonda önemli bir role sahip olan ,folat metabolizmasınındüzenlenmesinde anahtar bir enzimdir. Bu çalısmada meme kanseri veMTHFR gen polimorfizmi arasındaki iliski arastırılmıstır.Periferik kandan DNA izolasyonları yapılan 199 hasta ve 195kontrol , PCR-RFLP yöntemi kullanılarak incelenmistir. Elde edilensonuçların degerlendirilmesinde ; hasta ve kontrollerin genotip ve allelsıklıkları karsılastırılmıs istatistiksel olarak anlamlı bir farkbulunmamıstır. Genotiplerden, CC+CT'ye karsılık TT veya CC'yekarsılık CT+TT karsılastırıldıgında da istatistiksel olarak bir iliskitesbit edilmedi. Ancak meme kanserinin bir tipi olan infiltratif duktalkarsinomlu hastalarda MTHFR geni C677T polimorfizmi arasındaistatiksel olarak anlamlı bir iliski oldugu gözlemlenmistir. (p=0.038)statiksel olarak anlamlı olmasada TT homozigot bireylere, hastapopülasyonunda kontrollere göre daha sık rastlanmıstır.
dc.description.abstractABSTRACTBreast cancer is the most common cancer in women according to data from theMinistry of Health of Turkey in 2006.According to data in 2011,one of everyeight women in the United States will be faced with risk of breast cancer in alifetime.MTHFR is an enzyme that reduces 5,10-methylenetetrahydrofolate to 5-methyltetrahydrofolate, a carbon donor for the homocysteine to methionineconversion.MTHFR is a key enzyme that regulates folate metabolism which hasan important role in DNA synthesis, DNA repair and methylation.In thisstudy,associations of MTHFR C677T polymorphism with breast cancer havebeen investigated.199 patients and 195 controls were analyzed using PCR-RFLP method.GenomicDNA isolation was performed from peripheral blood.Allele and genotypefrequencies of patients and controls were compared to evaluate the results whichwere obtained and no statistically significant differences were found.Also,whenCC+CT with TT genotypes and CT+TT with CC genotypes were compared,nostatistically significant association was observed.However,it was observed astatistically significant association between MTHFR gene C677T polymorphismand infiltrative ductal carcinoma which is a type of breast cancer. (p=0.038)TThomozygous individuals were encountered more frequently in patients thancontrols although it was not statistically significant.en_US
